EN-US: This work aimed to evaluate the adsorption isotherms of roasted and grinded coffee, in different blends and storage temperatures. Crude grain coffee of two species were used: arabica (Coffea arabica) and robusta(Coffeacanephora), dehulled and dried. Grain were submitted to a selection to eliminate deteriorated grain, damaged and bored, to obtain a homogeneous raw material, without defects. Grain coffee from both species, after triage, were sent to roasting process at medium light level (Agtron SCAA#65).After roasting process, grain wasprocessed in a Mahlkönig mill, at average particle size (0.84 m). Blends were made in the following proportions of arabica and robusta: 80:20; 60:40 and 40:60.Samples were stored in BOD chambers for 6 months and kept at temperatures of 10 and 30 ºC, with initial moisture content of 2.0 ± 0.3 % (d.b.). At the beginning of storage (time zero) and at 30, 60, 120 and 180 days of storage, moisture content and water activity were determined. Several mathematical models, frequently used to represent the agricultural products hygroscopicity, were fitted to experimental data of sorption. Values of water activity and moisture content varied between 0.1912 and 0.5160; 2.20 and 5.51 % (d.b.), respectively. Modified Oswin model was the one that best represented the adsorption of coffee blends. | PT-BR: Este trabalho teve como objetivo avaliar as isotermas de adsorção de café torrado e moído, em diferentes formulações de blends e temperaturas de armazenamento. Foram utilizados grãos de café cru de duas espécies: arábica (Coffeaarabica) e robusta (Coffeacanephora), descascados e secos. Os grãos foram submetidos a uma triagem para eliminar os grãos deteriorados, danificados e brocados, a fim de se obter uma matéria-prima homogênea e isenta de defeitos. Os grãos de café de ambas as espécies, após a triagem, foram encaminhados para o processo de torrefação ao nível média clara (Agtron SCAA#65). Após o processo de torrefação, os grãos foram processados em moinho Mahlkönig, na granulometria média (0,84 mm). A formulação dos blends se deu nas seguintes proporções de arábica e robusta: 80:20; 60:40 e 40:60. As amostras foram armazenadas em câmaras BOD durante 6 meses e mantidos às temperaturas de 10 e 30 ºC, com teor de água inicial de cerca de 2,0 ± 0,3 % (b.s.). No início do armazenamento (tempo zero) e aos 30, 60, 120 e 180 dias de armazenamento foram determinadas o teor de água e a atividade de água. Aos dados experimentais de sorção foram ajustados diversos modelos matemáticos frequentemente utilizados para representação da higroscopicidade de produtos agrícolas. Os valores de atividade de água e teor de água variaram entre 0,1912 e 0,5160; 2,20 e 5,51 % (b.s.), respectivamente. O modelo de Oswin modificado foi o que melhor representou a adsorção de blends de café. Babesia bovis is a tick-borne pathogen that remains an important constraint for the development of cattle industries in tropical and subtropical regions of the world. Effective control can be achieved by vaccination with live attenuated phenotypes of the parasite. However, these phenotypes have a number of drawbacks, which justifies the search for new, more efficient immunogens based mainly on recombinant protein technology. In the present paper, ribosomal phosphoprotein P0 from a Brazilian isolate of B. bovis was produced and evaluated with regard to conservation and antigenicity. The protein sequence displayed high conservation between different Brazilian isolates of B. bovis and several Apicomplexa parasites such as Theileria, Neospora and Toxoplasma. IgG from cattle experimentally and naturally infected with B. bovis as well as IgG1 and IgG2 from naturally infected cattle reacted with the recombinant protein. IgG from cattle experimentally infected with Babesia bigemina cross-reacted with B. bovis recombinant P0. These characteristics suggest that P0 is a potential antigen for recombinant vaccine preparations against bovine babesiosis. The aims of the present paper were to analyse the genetic conservation of the P0 gene among Brazilian isolates of B. bovis and produce recombinant P0 (rPO) from a Brazilian isolate and evaluate its antigenicity with sera from cattle naturally or experimentally infected with B. bovis and sera from cattle experimentally infected with B. bigemina in an enzyme-linked immunosorbent assay (ELISA).
