Background: The pathogenesis related protein PR10 (TcPR-10), obtained from the Theobroma cacao-Moniliophthora perniciosa interaction library, presents antifungal activity against M. perniciosa and acts in vitro as a ribonuclease. However, despite its biotechnological potential, the TcPR-10 has the P-loop motif similar to those of some allergenic proteins such as Bet v 1 (Betula verrucosa) and Pru av 1 (Prunus avium). The insertion of mutations in this motif can produce proteins with reduced allergenic power. The objective of the present work was to evaluate the allergenic potential of the wild type and mutant recombinant TcPR-10 using bioinformatics tools and immunological assays. Methodology/Principal Findings: Mutant substitutions (T10P, I30V, H45S) were inserted in the TcPR-10 gene by sitedirected mutagenesis, cloned into pET28a and expressed in Escherichia coli BL21(DE3) cells. Changes in molecular surface caused by the mutant substitutions was evaluated by comparative protein modeling using the three-dimensional structure of the major cherry allergen, Pru av 1 as a template. The immunological assays were carried out in 8?12 week old female BALB/c mice. The mice were sensitized with the proteins (wild type and mutants) via subcutaneous and challenged intranasal for induction of allergic airway inflammation.,Conclusions/Significance: We showed that the wild TcPR-10 protein has allergenic potential, whereas the insertion of mutations produced proteins with reduced capacity of IgE production and cellular infiltration in the lungs. On the other hand, in vitro assays show that the TcPR-10 mutants still present antifungal and ribonuclease activity against M. perniciosa RNA. In conclusion, the mutant proteins. Resumo: A fruticultura no Nordeste brasileiro vem se destacando e a região sul da Bahia acompanha esse desenvolvimento com agroindústrias sendo instaladas por toda região, o que vem gerando resíduos agroindustriais que podem ser aproveitados na dieta animal. O objetivo deste trabalho foi avaliar o valor nutritivo dos resíduos gerados pelas agroindústrias processadoras de polpas de frutas da região sul da Bahia para alimentação de ruminantes. Os resíduos de frutas (caju, goiaba, graviola, manga e umbu) foram avaliados quanto a composição bromatológica, digestibilidade in vitro e cinética de fermentação ruminal pela técnica in vitro semi-automática de produção de gases. Os teores de matéria seca variaram de 12,4% (Graviola) a 42,92% (umbu) e os de proteína bruta de 4,95% (Manga) e 10,7 (Graviola). Os teores de parede celular foram elevados, destacando-se o umbu com os valores mais elevados de fibra detergente neutro (FDN, 88%) e fibra detergente ácido (FDA, 74,8%) e a casca da Manga com os menores valores, 23,7% de FDN e 14,5% de FDA. Quanto aos valores de digestibilidade in vitro e cinética de fermentação ruminal, destacou-se a casca de Manga, Enquanto os resíduos de Goiaba e Umbu foram inferiores aos demais. Os resíduos de frutas estudados apresentaram potencial para serem utilizados como alternativa na alimentação de ruminantes, porém são necessários estudos envolvendo resposta animal.
[Chemical composition and ruminal fermentation kinetics of fruit by-products in south Bahia - Brazil]
Abstract - The fruit production in the northeast of Brazil has been improved, especially in Bahia state where fruit companies were installed. During the fruit processing, several by-products are produced which could be used for animal nutrition. The objective of this work was to evaluate the nutritional value of fruit by-products generated by fruit companies on the south of Bahia state. The fruit by-products, Cashew (Anacardium occidentale), Guava (Psidium guajava), Soursop (Annona muricata L.), Mango (Mangifera indica) and Umbu (Spondias tuberosa) were analyzed in terms of chemical composition and by the in vitro gas production technique.
The dry matter content ranged from 12.4% (soursop) to 42.92% (Umbu) and 4.95% (Mango) to 10.7 (Soursop) for crude protein. The cell wall contents were very high; the Umbu neutral fiber detergent (FDN) was 88.0% and acid detergent fiber (FDA) 74.8% and the Mango peel promoted the lowest values (23.7% for FDN and 14.5% for FDA). Mango peel demonstrated the highest potential for ruminal digestibility and kinetics. Guava and Umbu residues demonstrated the lowest nutritional value.
The residues of fruits showed future as alternative feeds for ruminants, however, in vivo studies are needed.
