four isolates of passion fruit woodiness virus (PWV) were collected from Passiflora edulis f. flavicarpa, P. edulis, P.alata and Phaseolus vulgaris in the Federal District and Cristalina, Goias State, and characterized.Cytoplasmic inclusions were seen at the light microscope which were later characterized as pinwheel nad lamellar inclusions as seenin the electron microscope. Filamentous virus particles, typical of potyviruses, were seen in leaf dip preparations from infected samples. In many leaf parenchyma cells of infected plants some chloroplasts appeared in different degress of disorganization. Serological reactions using antiserum against a Brazilian isolate of PWV resulted positive.They were inoculated onto several test plants, mostly leguminous, in anattempt to distinguish these isolates. Although some slight differences were seen in the intensity of the symptoms, no distinction could be found among the isolates. They infrected most cultivars of Phaseolus vulgaris but not P. angularis, Vigna radiata and V. unguiculata. P. vulgaris cv. Rico-23, Rosinha and Preto-153 were the most susceptible with very severe symptoms; cv. Jalo was also infected, but cv.Top Crop and Dubbele Witte were not infected. |
