Ninety-eight commercial crossbred lambs, average weight 55.1 kg (+- 4.95), were used to evaluate bioelectrical impedance analysis (BIA) as a prediction method for fat-free tissue from live lambs and lamb carcasses. Lambs were transported to the abattoir, restricted from feed for 10 h, weighed (LWt), and measured for body resistance (Rs, ohms), body reactance (Xc, ohms), and distance between detector terminals (L, cm). Following slaughter, hot carcasses were weighed (HCWt; average 31.4 +- 2.8 kg) and impedance measurements of Rs, Xc, and L were recorded from readings on the dorsal and lateral Bides of the carcasses. Temperatures (average = 39.2°C) were recorded. Carcasses were chilled for 24 h. Cold carcass weights (CWt; average 31.1 :t 2.8 kg) and temperatures (average = 1.0°C) were recorded and BIA measurements were repeated. Carcasses were split down the midline and right Bides were ground three times and sampled for chemical analysis of fat, moisture, ash, and protein. Fat-free mass (FFM) was calculated as [Wt - (Wt x %Fat)]. Fat-free soft tissue (FFST) was calculated as the sum of carcass chemical protein and carcass water. Regression equations predicting fat-free tissue for the live lambs (FFMlive and FFSTlive), hot carcasses (FFMhot and FFSThot), and cold carcasses (FFMcold and FFSTcold) were developed. The independent variables LWt, Rs, and Xc accounted for 77.7% (residual mean square error [RMSE] = 1.97 kg) of the variation in FFMlive and 78.6% (RMSE = 1.78 kg) of the variability ofFFSTlive. Hot or CWt, Rs, Xc, L, and temperature detected 77.9% of the variation for FFMhot (RMSE = 1.04) and FFMcold (RMSE = 1.04) and 79.1 (RMSE = .92 kg) and 77.6% (RMSE = .95 kg) of the variation in FFSThot and FFST cold, respectively. The results show that bioelectrical impedance bas potential as a means of predicting the fat-free component of market-weight lambs and lamb carcasses. This study evaluated the effect of reduced water intake on survival, apoptosis and immunoexpression of leptin in sheep preantral follicles, activation of primordial follicles, serum levels of leptin, estradiol (E2) and progesterone (P4), and in vitro maturation (IVM) of oocytes antral follicles, as well evaluated the effects of leptin on in vitro culture of secondary follicles isolated these animals. Ewes (n = 32) were divided into four groups: water ad libitum (Control ? 100%), 80%; 60% and 40% of ad libitum intake. Blood was collected to determine, leptin, E2 and P4, before and after experiment. After the slaughter, ovarian cortex was used to histological and immunohistochemistry analysis and oocytes IVM. Moreover, isolated secondary follicles were cultured in vitro for 12 days in control medium (α-MEM+) or α-MEM+ with 10 or 25 ng/mL leptin. The reduction of water intake caused a linear decreasing effect on the percentages of normal preantral follicles, especially of primordial (P < 0.05), increased the apoptosis (P < 0.05) and decreased leptin expression in preantral follicles. The treatment with 60% of water intake showed greater total growth rate of isolated secondary follicles cultured with 25 ng/L leptin (P < 0.05), compared to those cultured in α-MEM+ . In conclusion, reduced water intake impaired the number of normal sheep preantral follicles, especially of primordial follicles, increased apoptosis and decreased leptin expression in preantral follicles.
