Forty-four varieties of the world cassava collection (Manihot esculenta Crantz) held in vitro at CIAT since 1979 with subculture period between 7.5 to 18.4 months, were examined for their responses to the presence of silver nitrate, an ethylene inhibitor. In order to improve the traditional method of in vitro conservation of clones by reducing growth rate and increasing period of conservation, the classical formulation of medium was supplemented with various concentrations of silver nitrate (23.54uM, 47.06uM, 58.85uM, 70.62uM, 82.40uM). Eighteen months after initiating this study, all concentrations tested resulted in a major reduction of growth as related to the control. Best results were observed with concentrations between 58.85uM and 82.40uM, having all plants shorter internodes, a lower rete of defoliation, and a higher percentage of survival. |
