The storage proteins of soybean characteristically sediment in sucrose density gradients with coefficients of aproximately 7S (conglycinin) and 11S (glycinin). With sodium dodecyl sulfate and Beta-mercaptoethanol treatment the 7S protein breaks down into at least three polypeptide subunits, while the 11S protein breaks into four basic and four acidic subunits. Messenger RNAs have been isolated from polyribosomes taken from developing soybean seeds and after affinity chromatography on oligo dT-cellulose and sucrose density gradient centrifugation (under denaturing conditions) where translated in a wheat germ cellfree protein synthesizing system. Messenger RNA activities ranged from approximately 10S to 24S. Some, but not all, of the polypeptides synthesized invitro had electrophoretic mobilities identical with some of the subunits of the storage proteins. Preliminary evidence from tryptic fragment analysis suggested that the basic subunits of the 11S protein are synthesized in vitro; other products have not been comparably characterized. Polyacrylamide gel electrophoretic analysis of the poly(A) + RNAs showed a widely heterodisperse population of mRNAs with no predominating species. |
