Sementes de café (Coffea arabica L.) são classificadas como intermediárias em relação à tolerância à dessecação, são sensíveis à desidratação até certo grau de umidade, e tem sua armazenabilidade reduzida, o que tem sido um desafio para os produtores de sementes e programas de melhoramento. Análises ultraestruturais das membranas e da parede celular do endosperma e embrião podem contribuir para a compreensão dos processos de perda de qualidade durante a secagem e armazenamento das sementes de café. Assim, objetivou-se neste trabalho analisar as alterações ultraestruturais em sementes de café secadas até diferentes teores de água. Foram utilizadas sementes da safra 2012/2013, da espécie Coffea arabica L., cultivar Catuaí Amarelo IAC 62. As sementes foram submetidas à secagem rápida, em sílica gel, até atingirem os teores de água de interesse, de 40, 20 e 5% de umidade (base úmida). Após secagem, as sementes foram aleatoriamente amostradas e fragmentos do endosperma aderidos ao embrião foram preparados para a microscopia eletrônica de varredura, registrando-se diversas eletromicrografias. Foram avaliadas as alterações na membrana plasmática da célula do endosperma e do embrião das sementes de café. A secagem interfere na resistência e estrutura da parede celular e no conteúdo celular do endosperma e do embrião das sementes. As células das sementes de café úmidas apresentam-se túrgidas e o conteúdo interno celular intacto e sem contração. Observou-se comprometimento das estruturas celulares nas sementes de café com o progresso da secagem, sendo que as sementes com 5% de teor de água são as mais prejudicadas. a b s t r a c t The main objective of this study was to verify genetic trends for milk production (MY305) and age at first calving (AFC). Were also considered levels of inbreeding practiced in the Brazilian dual-purpose Guzera population (TPOP) comparing it with the same parameters estimated for two sub-populations derived from the reference (TPOP): female donors submitted to ovum-pick up (DPOP) and in vitro embryos produced (EPOP) between 2003 and 2013. Estimated breeding values (EBV) and inbreeding coefficients (F) were regressed by the year of birth (or year of in vitro fertilization) of each animal or embryo in order to obtain annual trends for these parameters separately for each of the three populations studied. A positive quadratic (b2 ¼ þ0.000075) effect was detected for the F values in TPOP. Both DPOP and EPOP showed positive linear coefficients (b1), respectively, þ0.00084 (P < 0.001) and þ0.00024 (P > 0.05). Annual mean F for EPOP was higher than TPOP and DPOP through the time series studied. The frequency of individuals with more than 7% F was higher in DPOP. Genetic trends for AFC were 0.187 days/year (P > 0.05); 0.557 days/year (P < 0.05) and 1.48 days/year (P < 0.05), respectively for TPOP; DPOP and EPOP. Genetic trends for MY305 were þ6.75 kg/year (P < 0.001); þ8.2 kg/year (P < 0.001) and þ10.5 kg/year (P < 0.05), respectively for TPOP; DPOP and EPOP. For both traits analyzed, EPOP showed the highest regression coefficients, which confirms a higher selection pressure and lower generation intervals previously expected from this sub-population. Results reported in the present study suggest that mean F is increasing in the Guzera population. Efforts for controlling inbred mating on in vitro fertilization should be considered, as the presence of a bottleneck effect seems to be getting shape on DPOP and EPOP.
