The CiLV-C genome was completely sequenced and this led to the possibility to express the p29, the putative capsid protein, in vitro, and produce a highly specific polyclonal antibody against it. This antibody was used in immunogold labeling assays in citrus leaf lesions caused by CiLV-C. Intense labeling was observed on the viroplasm while a more discrete labeling was noticed on the presumed virions. No significant labeling was observed on other cell components or in the uninoculated healthy control leaves. Similar assays using tissues infected with other cytoplasmic type of viruses transmitted by Brevipalpus mites as Passion fruit green spot, Schefflera ringspot, Salvia green spot, Hibiscus green spot and Solanum violaefolium ringspot did not result in labeling on the viroplasm or presumed virions indicating that they are distinct from CiLV-C. This antibody also permitted the detection of CiLV-C by the tissue blot method. |
