Abstract: Cassava is a highly versatile crop in the tropic and subtropical regions which can grow under conditions such as poor soil nutrients, high acid soil and drought, where other crops like maize are not suitable. In these regions cassava is an important calorie source for human and animal. One of the major problems limiting the production and distribution of cassava, is the rapid deterioration (PD) from 24 hours to 48 hours after harvest depending on the genotype. Preliminary research indicates that PD is an incomplete wound process involving de novo gene expression. However, the biochemical pathway and molecular mechanism of PD are poorly understood. Our aim is to try to identify and isolate genes and proteins involved in PD. Based on a new method of differential cDNA diplay called cDNA-AFLP, around 90 transcript derived fragments (TDF) have been isolated in order to establish a catalogue of differentially expreseed genes during the process of PD in cassava tubers. A subset of the TDFs suggest possible linkage to Pd. From the data we hope to gain more insight into the molecular basis of PD in cassava. An overview of the material used and the genes isolated and identified will be presented. |
