The production of friable embryogenic tissues is a prerequisite for the recovery of cassava plants genetically transformed by particle bombardment. Here we report the production of friable embryogenic callus from fourteen geographically diverse cultivars of cassava. Plant regeneration has been achieved from seven of these, although the frequency at which this was achieved is cultivar dependent. Genetic transformation experiments have resulted in the recovery of transgenic plants from cvs. TMS 60444, Bonoua Rouge and M. Col 1505 containing the uidA marker gene, the coat protein gene from cassava common mosaic virus and the African cassava mosaic virus replicase gene. This demonstrates that the production of transgenic cassava with a range of transgenes is becoming a routine and reliable process, and that attention can now turn to examining the phenotype of transgenic cassava plants. |
