The effect of capsid protein heterogeneity on various virus properties in purified turnip mosaic virus was investigated. Polyacrylamide-gel electrophoresis in the presence of sodium dodecyl (SDS) revealed three distinct capsid protein species, designated as fast form (MW 27,000), intermediate form (MW 29,000), slow form (MW 36,000). Virus with capsid protein in the fast or intermediate form sedimented about 12.3 S slower and had a buoyant density in CsCl about 0.01 g/cm3 greater than slow-form capsid containing virus. Immunodiffusion tests in the presence of SDS revealed that slow-form capsid protein was serologically distinct from fast or intermediate capsid forms. Little effect was detected in ultraviolet-absorption spectra virus containing either of the three capsid forms. |
