The objective of this investigation was to assess the use of a mutant form of the green fluorescence protein as a reporter gene in cassava. Sterile leaf explants were plated onto the primary somatic embryogenesis induction medium. After 20 days embryogenic callus was plated onto the secondary embryogenesis proliferation medium. Agrobacterium tumefaciens strain LBA4404 containing binary plasmid XC-1 blue encoding the green fluorescence protein, variant mgfp5, was used to transform secondary somatic embryogenic cells. Callus and embryos were examined after 7, 14 and 28 days following transformation for GFP activity using the confocal microscope (Zeiss inverted microscope LSM410). In the transgenic cassava embryo GFP expression was found predominantly in the pllasmalemma and nuclear membranes. |
