Asian Soybean Rust (ASR) is caused by the fungus Phakopsora pachyrhizi. Outbreaks of ASR have now occurred in all major soybean-producing countries including the United States. Thus far, only four genes (Rpp1, Rpp2, Rpp3 and Rpp4) conferring resistance to ASR have been identified. In previous work, Silva et al. (2007) mapped the Rpp2 resistance gene to a region on soybean linkage group J. Three SSR markers identified by Silva et al., were used to screen the Iowa State University/USDA-ARS "Williams 82" BAC library, resulting in three initial BAC contigs. To expand and coalesce the contigs, primers developed from BAC-end sequences were used to rescreen the BAC library. Concurrently, skim sequencing of BACs from within the region confirmed the presence of sequences with homology to plant disease resistance genes. While the ?Williams 82? genotype does not contain the Rpp2 resistance gene, the genomic resources developed with this genotype make it is an essential tool for eventual cloning of the Rpp2 gene. Skim sequence reads are currently being used to develop new SSR markers that will further delimit the region conferring resistance. |
