Citrus leprosis (CL), an important disease, which occurs mainly in Brazil and Argentina, is transmitted by mites of the genus Brevipalpus. Limited evidence suggests that CL is caused by virus. We report mechanical transmission of CL from citrus to citrus and three berbaceous testplants (Chenopodium amaranticolor, C. guinoa and Gemphrena globosa), with production of only local lesions, in all cases. Reaction times were 5 to 7 days for the two Chenopodium species, 14 to 16 days for G.globosa and 20 to 24 for citrus.In naturally infected citrus tissues and in experimentally obtained local lesions, non-enveloped rhabdovirus-like particles 120-130 X 50-55 nm were observed within membranous vesicles of the endoplasmic reticulum. Sometimes the particles reached 300nmin length. Dense masses (viroplasms), and vesicles 80 to 100 nm in diameter, associated with the tonoplast, were also observed. The cytopathology was not identical with that previously reported for CL. Mechanical transmissibility, the finding of virus-like particles in infected tissues, and the cytopathology eliminate the possibility of CL being caused by mite salivary toxins, and open the way to the identification of the causal agent, which is almost certINLY A VIRUS. |
