Abstract: Since its foundation in 1974, CENARGEN has actively stored plant genetic resources for food and agriculture. Among a number of conservation modalities, tissue culture stands out as a reliable long-term conservation method. The initial aims of the tissue culture laboratory since its establishment in 1978, were in vitro germplasm exchange and the release of virus-free clones. Later on, the scope of the unit was expanded to cover in vitro conservation of species. This move antecipated the recommendation of former International Board of Plant Genetic Resources (IBPGR) Advisory Committee for the instalation of in vitro storage facillitis in seed gene banks. The unit started the in vitro conservation with 150 accessions of cassava mostly received from the Centro International de Agricultura Tropical (CIAT). During the early 1980's an active and massive interchange of in vitro cassava germplasm took place between CENARGEN and CIAT. Meanwhile a great success was achieved with the increasing mastering of cost-effective slowgrowth media techniques. Among other achievements in this area, the reduction of explants growth obtained as a result of diminishing temperatures in the growth chambers is worth the record. The unit works with a subculturing average replacement is of 10 months. CENARGEN's in vitro collection of Manihot species is kept at 20+2°C and the inventory consists of 1220 M. esculenta accessions besides 75 wild accessions belonging to 20 species. |
