Micropropagated P. pinaster shoots were cultured for 16 days on a basal medium containing NAA and radiolabelled nitrate (3.3 mM nitrogen), supplemented with glutamine (2 mM nitrogen, control medium) and/or K-glutamate (2 mM nitrogen). Culture on the control medium resulted in optimal rooting, whereas rooting was inhibited by the presence of glutamate. Study of 15NO3- behaviour in shoots cultured on the control medium showed a marked increase in 15N incorporation during the second half of the treatment period. The majority of the nitrogen incorporated from the medium was accumulated into protein in the rooting zone. Autoradiography of plants treated with (3H)-thymidine showed strong DNA synthesis in the rooting zone, which increased up to day 8. Addition of glutamate resulted in a marked slowdown of nitrate uptake and a decrease in protein synthesis. |
