RESUMO: Os rizóbios representam um grupo notório de bactérias fixadoras de nitrogênio (BFN) pela sua habilidade em disponibilizar N às leguminosas, reduzindo o uso de fertilizantes nitrogenados. Dessa forma, estudos que visem identificar, caracterizar e avaliar aspectos ligados à eficiência simbiótica de rizóbios são de grande importância. O objetivo deste trabalho foi caracterizar taxonômica e filogeneticamente estirpes de rizóbios provenientes dos Cerrados e da Mata Atlântica, dois biomas importantes ecologicamente e economicamente para o país. Foram realizados três estudos, para os quais foram feitas análises de caracterização molecular e análises filogenéticas. No Estudo 1, foi realizada uma análise polifásica em quatro estirpes de Rhizobium, que apresentaram características diferenciadas em estudos anteriores. Os resultados obtidos confirmaram que este grupo de estirpes representava uma nova espécie, a qual foi denominada Rhizobium paranaense, e a PRF 35 T foi definida como estirpe tipo para esta espécie. No Estudo 2 foi feita uma caracterização genética de 181 isolados obtidos em solos não-perturbados dos Cerrados, utilizando-se o feijoeiro comum (Phaseolus vulgaris) como planta-isca. este estudo, a espécie Paraburkolderia nodosa foi predominante e duas estirpes apresentaram alta taxas de fixação de N2. No estudo 3, 427 estirpes provenientes de solos da Mata Atlântica foram isoladas e caracterizadas utilizando-se Mimosa pudica e Phaseolus vulgaris como plantas-isca. O gênero Paraburkholderia foi predominante na maioria das amostras, sendo que seis possíveis novas espécies foram identificadas. A análise canônica foi significativa para o pH do solo, indicando que o mesmo influenciou na diversidade de bactérias nessas amostras. Os três estudos elucidaram que ainda existe muito a ser descoberto em relação à diversidade de rizóbios nos trópicos, sendo que a caracterização destes microrganismos contribui não apenas para identificar novas estirpes, mas também para explorar suas propriedades biotecnológicas. ABSTRACT: Rhizobia represent a remarkable group of simbiotic nitrogen-fixing bacteria (NFB) due to their ability of providing N for leguminous plants, reducing the use of nitrogen - fertilizers. In this way, studies that intend to identify, characterize and evaluate aspects related to the symbioses legume - rhizobia are of great importance. The objective of this study was to characterize taxonomically and phylogenetically rhizobial strains that were isolated from the Cerrado and the Mata Atlântica soils, two important biomes for the economy and ecology of Brazil. Three studies were conducted using molecular and phylogenetic analyses for bacteria characterization. In the first study, a polyphasic analysis was performed with four Rhizobium strains that presented different features in previous studies. The results confirmed that this group of strains represents a new species, which was nominated Rhizobium paranaense, and PRF 35 T was defined as the type strain forth e species. In the second study, a genetic characterization was performed with 181 isolates from undistu rbed soils of the Brazilian Cerrado, using common bean (Phaseolus vulgaris) as trap-host plant. In this study, the predominant species was Paraburkolderia nodosa , and two strains presented high rates of N2 fixation. In the third study, 427 strains from the Mata Atlântica soils were isolated using Mimosa pudica and Phaseolus vulgaris as trap-host plants. Paraburkholderia was the predominant genus in most soil samples, and six potential new species were identified. The canonic analysis was significant for soil pH, indicating that it influenced bacterial diversity. The three studies highlighted that there is still much to be discovered about rhizobial diversity in the tropics, and the characterization of the rhizobia diversity can contribute not only for the identification of new species, but also to explore their biotechnological properties.
