Cultivated in Liquid Medium ResumoAbstract: The exploration of microorganisms as a source of biologically active substances have been performed since the discovery of the first fungal metabolite of great medical importance, the penicillin. Thus, the main objective of this study was to evaluate the chemical components in the extract of mycelium of Tyromyces pulcherrimus, and to evaluate the potential in vitro antibacterial, antioxidant and inhibitory activities of alpha-glucosidase and acetylcholinesterase enzymes. The classes of secondary metabolites detected by thin layer chromatography (TLC) were flavonoids, phenols and lipids. An analysis by high-performance liquid chromatography coupled with mass spectrometry (HPLC-ESI-MS/ MS) comparing to the 47 standards of phenolic compounds, led to the identification of ellagic acid (4.55 ± 0.15 μg g-1). The extract showed weak antibacterial activity for Pseudomonas aeruginosa (ATCC 27853) and showed no antibacterial activity against Escherichia coli (ATCC 25922) and Staphylococcus aureus (ATCC 25923). In anticholinesterase activity, 99 ± 0.1% of the enzyme was inhibited at a concentration of 1 mg mL-1. At the same concentration, the sample showed weak antioxidant activity on the DPPH assay (18.18 ± 0.04%), and weak inhibitory activity of the enzyme alpha-glycosidase IC50 of 15.19 ± 5.52%. This study suggests that the anticholinesterase activity in this species is promising and may be involved with the compounds detected by TLC and mass spectrometry.A exploração dos microorganismos como fonte de substâncias biologicamente ativas tem sido realizada desde a descoberta do primeiro metabólito fúngico de importância na medicina, a penicilina. O objetivo principal deste estudo foi avaliar os componentes químicos presentes no extrato do micélio fúngico da espécie Tyromyces pulcherrimus, e avaliar as potenciais atividades antibacteriana, antioxidante e inibitória das enzimas alfa-glicosidase e acetilcolinesterase in vitro presentes na espécie. As classes de metabólitos secundários detectados por cromatografia em camada delgada (CCD) foram flavonoides, fenóis e lipídios. Uma análise por cromatografia líquida de alta eficiência acoplada a espectrometria de massas (HPLC-ESI-MS/MS), em comparação a 47 padrões de compostos fenólicos, permitiu a detecção do ácido elágico (4,55 ± 0,15 μg g-1). O extrato mostrou fraca atividade antibacteriana para Pseudomonas aeruginosa (ATCC 27853) e não mostrou atividade antibacteriana frente à Escherichia coli (ATCC 25922) e Staphylococcus aureus (ATCC 25923). Na atividade anticolinesterásica, houve inibição de 99 ± 0,1% da enzima na concentração de 1 mg mL-1. Nesta mesma concentração, a amostra apresentou fraca atividade antioxidante no ensaio com o radical livre DPPH (18,18 ± 0,04%) e fraca atividade inibitória da enzima alfa-glicosidase (15,19 ± 5,52%). Este estudo sugere que a atividade anticolinesterásica nesta espécie é promissora e pode estar envolvida com os compostos detectados por CCD e espectrometria de massas. MenosRev.
