In peach, pollen sterility, expressed as ab-sence of pollen in the anthers, segregates as an undesired trait in breeding programs. Pollen fertility screening inprogenies is not a common practice mainly because itdoes not affect fruit set since cross-pollination is fre-quent. It is also a time-consuming activity that coincideswith the busy pollination season. Segregation for thistrait could be avoided by using molecular markers toidentify appropriate parents or male sterile plants forearly culling in progenies expected to segregate, thusincreasing breeding efficiency. In peach, pollen sterilityis determined by a recessive allele in homozygosis of themajor gene,Ps/ps, located on chromosome 6. In thiswork, using a conventional mapping approach com-bined with bulked segregant analysis usingresequencing data, we fine mappedPsto a region ofalmost 160 kb and developed molecular markers formarker-assisted breeding. These markers were validatedin plant materials from three peach breeding programs,including progenies, advanced selections, and cultivars,allowing us to determine that the frequency of thepsallele is high (0.23) and also to infer the genotypes of alarge collection of cultivars and advanced breedinglines. |
