This research work was carried out at the Biotechnology Lab. from INIVIT, in order to establish a regeneration system of somatic embryogenesis for mass plant production and transgenesis in cassava (Manihot esculenta Crantz). The following clones were used: `Senorita', `CEMS 74-725' and `CEMSA 74-6329' (Cuban genotypes) and `CMC-76' and `CMC-40' clones from our gene banks. The effects of different 2,4-D (Dichlorophenoxy-acetic acid) concentrations and explant types to obtain embryogeniccalli were tested in `Murashige and Skoog' medium. The Temporary Immersion System was used to increase efficiency of the conversion process from embryos to plants. Plants regenerated by somatic embryogenesis and meristem cultures plants propagated by somatic embryogenesis and meristem cultures during hardening and field stages were compared with plants propagated through traditional systems. |
