Abstract: This study aimed to evaluate two hormonal protocols for synchronization of follicular wave emergence on in vivo embryo production in Santa Ines sheep under tropical conditions. Twenty two nulliparous Santa Ines sheep, kept on intensive system, were used. Group one (GT, n = 10) received an intravaginal implant containing 0.33 g of progesterone (Eazi-Breed CIDR® Sheep and Goats, Zoetis Ltda, São Paulo, Brazil) for all period of hormonal protocol and 0.24 mg of cloprostenol (Estron®, Agener Union, São Paulo, Brazil) i.m.. Group two (GEm; n = 12) received the synchronization of the follicular wave emergence as proposed by Balaro et al. (2016). The superovulation started after 56 and 80 hours, from GT and GEm, respectively. For both groups, 200 mg of FSHp/per animal were administered (Folltropin®, National Pharmaceutical Chemistry Union S/A, São Paulo, Brazil) in six decreasing doses (25%/25%, 15%/15% and 10%/10%) every 12 hours. In GEm, at the first FSHp dose, an intravaginal sponge impregnated with 60 mg of medroxyprogesterone acetate (Progespon®, Schering Plough, São Paulo, Brazil) was inserted and at the fifth dose, ewes also received 0.24 mg of cloprostenol (Estron®, Agener Union, São Paulo, Brazil). The intravaginal implant (GT) or sponge (GEm) was taken at the last FSHp dose. Subsequently, estrus detection and mating occurred every 12 hours. Seven days after the last FSH dose, embryos were collected by surgical uterine flushing. An ultrasound (Sonoscape S6®, Sonoscape, Yizhe Building, Yuquan Road Shenzhen, China) equipment coupled to a 7.5 MHz linear transducer (transrectal) was used to assess the follicular population on the progestagen insertion (D0), in the first superovulatory dose and for CL count immediately before embryo collection. Furthermore, regardless of the occurrence of premature regression of CL (PRCL), all ewes were collected. Normal and non-normal parametric data were evaluated by t-test and MannWhitney, respectively. Frequency data were evaluated by Fisher's exact test. It was considered as significant when P < 0.05. No differences (P > 0.05) were detected, between GT and GEm, in the follicular population at the day of the progestagen insertion (7.8 ± 2.6 vs. 6.0 ± 1.6) and the first FSHp dose (8.1 ± 2.6 vs. 8.9 ± 2.3). The number of CL were also similar (P > 0.05) between G1 (6.9 ± 5.1) and G2 (7.1 ± 3.1). The number of animals with PRCL in GT was 60% (6/10), greater than the 8.3% (1/12) found in GEm (P < 0.05). The number of collected structures (6.6 ± 4.3 vs. 0.6 ± 0.7; P < 0.01) and viable embryos (4.6 ± 3.9 vs. 0.3 ± 0.5; P < 0.01) were greater in GEm compared to GT. The recovery rate also differed between groups, being greater in GEm than GT (75.6% vs. 8.1%; P < 0.01). The greater PRCL rate in GT probably contributed to the smaller number of viable embryos. Thus, it is suggested the appliance indicated the GEm protocol for in vivo embryo production in Santa Ines sheep under tropical conditions. [Sincronização da emergência folicular para aumentar a produção in vivo de embriões ovinos da raça Santa Inês]. Resumo: O estudo objetivou a avaliação de dois protocolos hormonais para a sincronização da emergência da onda folicular na produção de embriões em ovinos da raça Santa Inês sob clima tropical. Foram utilizadas 22 ovelhas nulíparas da raça Santa Inês mantidas sob regime intensivo. Os animais de um grupo receberam um tratamento tradicional (GT; n=10) um dispositivo intravaginal contendo 0,33g de progesterona (Eazi-Breed CIDR® Ovinos e Caprinos, Zoetis Ltda, São Paulo, Brasil) e 0,24 mg de clorprostenol (Estron®, Agener União, São Paulo, Brasil) por via intramuscular. As ovelhas do outro grupo tiveram a emergência da onda folicular sincronizada conforme proposto anteriormente (Balaro, M. Domestic Animal Endocrinology, v.54, p. 10-14, 2016) (GEm; n=12). A superovulação iniciou após 56 e 80 horas, no GT e GEm, respectivamente. Para ambos os grupos, foi utilizado 200 mg de FSHp (Folltropin®, União Química Farmacêutica Nacional S/A, São Paulo, Brasil) em seis doses decrescentes (25%/25%, 15%/15% e 10%/10%), a cada 12 horas. No GEm, no momento da aplicação da primeira dose de FSHp foi inserida uma esponja impregnada com 60 mg de acetato de medroxiprogesterona (Progespon®, Schering Plough, São Paulo, Brasil). Na aplicação da quinta dose de FSHp, as ovelhas também receberam 0,24 mg de clorprostenol (Estron®, Agener União, São Paulo, Brasil) e a esponja (GT) ou o implante intravaginal (GEm) foram retiradas ao administrar na última dose de FSHp. A detecção do estro e coberturas ocorreram, na sequência, a cada 12 horas. Após sete dias da última dose de FSH, os embriões foram coletados por meio da lavagem uterina cirúrgica. Utilizou-se o ultrassom (Sonoscape S6®, SonoScape, Yizhe Building, Yuquan Road, Shenzhen, China) acoplado a um transdutor linear de 7,5 MHz (via transretal) a fim de se avaliar a população folicular no dia do implante do progestágeno (D0), na primeira dose superovulatória e na contagem de CL antes da coleta de embriões. A ocorrência de regressão prematura de corpos lúteos (RPCL) também foi verificada. Os dados paramétricos normais e não-normais foram avaliados pelo teste t e Mann-Whitney, respectivamente. Dados de frequência foram avaliados pelo teste Exato de Fisher. Para ambos, considerou-se como significativo quando P<0,05. Não foram encontradas diferenças (P<0,05) na quantidade de folículos no dia da inserção do progestágeno (7,8 ± 2,6 vs. 6,0 ± 1,6) e na primeira dose de FSH (8,1 ± 2,6 vs. 8,9 ± 2,3). O número de CL contados no dia da coleta também não diferiu (P>0,05) entre GT (6,9 ± 5,1) e GEm (7,1 ± 3,1). O número de animais com RPCL no GT foi de 60% (6/10), superior (P<0,01) aos 8,3% (1/12) encontrados no GEm. O número de estruturas coletadas (6,6 ± 4,3 vs. 0,6 ± 0,7; P<0,01) e embriões viáveis (4,6 ± 3,9 vs. 0,3 ± 0,5; P<0,01) foram superiores no GEm comparado ao GT. A taxa de recuperação também diferiu entre os grupos experimentais, sendo superior no GEm (75,6% vs. 8,1%; P<0,01). A maior taxa de RPCL no GT contribuiu para o menor número de embriões viáveis obtidos. One of the most relevant issues in discussion worldwide nowadays is the concept of sustainability. However, sustainability assessment is a difficult task due to the complexity of factors involved in the natural world added to the human interference. In order to assess the sustainability of beef ranching in complex and uncertain tropical environment systems this paper describes a decision support system based on fuzzy rule-approach, the Sustainable Pantanal Ranch (SPR). This tool was built by a set of measurements and indicators integrated by fuzzy logic to evaluate the attributes of the three dimensions of sustainability. Indicators and decision rules, as well as scenario evaluations, were obtained from workshops involving multi-disciplinary team of experts. A Fuzzy Rule-Based System (FRBS) was developed to each attribute, dimension and general index. The essential parts of the FRBS are the knowledge database, rules and the inference engine. The FuzzyGen andWebFuzzy tools were developed to support the FRBS and both showed efficiency and low cost for digital applications. The results of each attribute, dimension and index were presented as radar graphs, showing the individual value (0e10) of each indicator. In the validation process using theWebFuzzy, different combinations of indicators were made for each attribute index to show the corresponding output, and which confirm the feasibility and usability of the tool. |
