The interest on immunosensors has been increasing because of their utility as specific, simple, rapid, label-free, direct detection techniques with small sample volumes compared with conventional immunoassays. For immunosensors, the immunologic materials are immobilized on a solid supporting material. The immobilized biomolecule retains their bioactivity and interact with their counterparts, and based on the electrochemical detection, the amount or concentration of analytes can be determined. One challenge related to this application is that the reaction of antibody and the antigen-immobilized surface is often insufficient to produce a large electrochemical signal change. It has been showed that the employment of nanoparticles in the building of modified electrodes helps the amplification of the transducing signals of biomolecules interactions, like antigen-antibody. The emergence of nanotechnology has opened new opportunities for the application of nanoparticles in analytical electrochemistry. Additionally, there has been considerable interest in applying immobilized biomolecules (proteins, enzymes, DNA) to specific assays. Among these applications, highly sensitive detection of proteins can be accomplished via antigen-based immunoassays, with the recombinant protein attached to a solid surface. The similar dimensions of nanoparticles and protein molecules enable the enormous electrochemical signal enhancement in a biosensing device. The nanoparticle-biomolecule assemblies provide the basis for ultrasensitive electrochemical biodetection as on impedimetric transducers. In the development of the immunosensor, object of this work, a nanostructured solid surface has been built with the ability to couple the MSP5, recombinant protein from Anaplasma marginale and its correspondent antibody. Screen-printed gold electrodes are used for this purpose and gold nanoparticles layers are mounted on them with the aim of amplifying the impedance detection, or electrochemical signal.
