Conteúdo:  Soybean (Glycine max (L.) Merr.) is a major crop in Kentuchy. Soybean mosaic virus (SMV), bean pod mottle virus (BPMV), and dual infection with SMV and BPMV present a significant potential threat to soybean production in the state. The specific goals of this study were to identify the primary sources of BPMV inoculum in soybean fields; to investigate the mechanisms underlying the synergistic interactions between SMV and BPMV; and to construct chimeric genes containg SMV coat protein (CP) sequence for the purpose of producing CP-mediated transgenic resistance to SMV in soybean. The significance of overwintering bean leaf beetles, vectors of BPMV, and seeds from infected soybean plants as sources of primary virus inoculum was assessed. Although BPMV antigen wasreadily detected by ELISA in regurgitant from naturally overwintered beatles, none of these beetles transmitted the virus to healthy soybean seedlings. An average of 6.6% of transmission, however, was observed when beetles were exposed to artificial overwintering conditions. No apparent structural changes in BPMV RNA could be related to the loss or decline of BPMV infectivity in overwintering beetles. On the other hand, evidence is presented to indicate that exposure of BPMV to beetle regurgitant led to proteolysis of the CP.
