Soybean Glycine max (L.) Merril.have proven to be extremely resistant to transformation. In Brazil, transformation of soybean had succeeded using particle bombardment (bio-ballistic). In this study, soybean transformation using Agrobacterium was optimized. Mature soybean seeds were soaked for 16h and the embryonic tip, cotyledonary node and hypocotyl segment were collected. These explants were inoculated with Agrobacterium tumefaciens strain EHA105, which contains the construction pGreenII0229 integrated with 35S:GUS:NOS cassette. The use of embryonic tips yielded a higher transformation frequency when compared to cotyledonary nodes and hypocotyls. Results showed that T-DNA transfer efficiency reached to approximately 80%, and transformed shoot primordia and leaf primordia were observed on shoot elongation medium. |
