Rams were experimentally infected by the conjonctival route with B. ovis when four months of age and the course of infection was followed over a period of 17 weeks. An improved selective medium permitted the isolation of B. ovis from the semen of 20 of 23 rams. Of the 20 excreting rams, ii had palpable lesions. Complement fixation and gel precipitation tests were performed with rough specific antigen prepared by heat-extraction of B. ovis. Sera taken every fortnight showed that the gel test was invariably positive if the complement fixation titer was I : 20 or higher and usually positive if the complement fixation titer was I : 10. Of 21 rams with bacteriological evidence of infection, 10 rams had serological reactions in both tests. This indicates the reliability of serological tests for diagnosis and suggests that the gel test, which is easier to perform, may be substituted for the complement fixation test. Allergic tests were performed by the palpebral route with B. melitensis protein antigen to see if the same allergen could be employed for both B. ovis and B. melitensis infection in sheep. Significant reactions were obtained in the rams but there was no correlation in individual animals between allergic reaction and degree of infection by other parameters. The allergic test should be used on a flock rather than on an individual basis. |
