A Tomato Spotted wilt virus (TSWV) isolate from the Canary Islands denoted C-TSWV, has been characterized. This isolate was collected from a diseased Capsicum annuum L. cv. Gedeon plant showing heavy mosaic, leaf distortion and stem necrosis. The host range of this isolate was similar to the CNPH, isolate studied in the Wageningen laboratory. High absorbance values were obtained with DAS: ELISA when C-TSWV was studied with those of CNPH, TSWV. In leaf-dip preparations from infected plants a large number of spheroidal particles with 80-100 nm in diameter were found. High yields of virus were obtained from infected Nicotiana rustica L. plants. The cytopathology of the C-TSWV was similar to that of other TSWV isotates. Virus particlesoccurred in clusters between membranes. Viroplasma with dense masses were found close to the clusters in the cytoplasm. This isolate induced in addition fibrous structures in the cytoplasm of some hosts. Colloidal gold conjgated to viral antibodies labelled both particles and dense masses but not the fibrous structures. |
