Banana is a crop constantly subjected to biotic and abiotic stresses. Stresses due to diseases and pests, drought and cold are responsible, alone or in combination, for a reduction of the yield and quality of banana fruit. To overcome these stresses, banana has developed mechanisms to perceive external signals and to respond with biochemical, physiological and morphological changes within the cell and throughout the entire plant. An understanding of the molecular mechanisms behind these responses may enable researchers to better understand the plant-stress interaction and to design more efficient ways of controlling the plant response to these stresses. The search and partial characterisation of 24 genes in the DATAMusa database of banana, which are orthologs to genes known to be related to biotic and abiotic stresses in other species, are described. The assessment of Musa acuminata Assembled EST Sequence (MaAES) positive for genes related to biotic and abiotic stresses was carried out through a keyword search in the DATAMusa database. Positive MaAESs were checked for cDNA clones, which potentially contain the entire coding region of the gene. The selected cDNA clones were fully sequenced and the consensus sequence submitted to ORF identification. The predicted amino-acid sequence was then submitted to Blast search against the SwissProt database as well as to allergenicity prediction based on protein sequence. An expression analysis was carried out for each of the 24 genes through RT-PCR using total RNA from banana leaves, peel, flower and roots as a template. Musa orthologs of pathogen-related protein 10, germin-like protein, ascorbate peroxidase, glutathione peroxidase, heat shock proteins, peroxiredoxin, superoxide dismutase, salt tolerance protein, lectin, selenium binding protein and 14-3-3 protein are among those identified and characterised in this study.
