Very rapidly frozen tumor cells can survive intracellular freezing. The cells thus frozen appear to be as translucent as intact unfrozen ones. In the present paper the behavior of rapidly frozen tumor cells is described at various temperatures above ?30°C using MTK sarcoma III, an ascites sarcoma of rat. Translucent frozen tumor cells were obtained by cooling the cells rapidly (cooling rate: 13–30°C/ sec) to temperatures between ?18°C and ?30°C. The results presented in this paper suggest that the frozen cells have many intracellular fine ice crystals which are too small to interfere with the ordinary transmitted light. These cells can survive freezing as long as they remain translucent in apparance, and can resume normal growth in the peritoneal cavity of rat, provided that they are thawed very rapidly (thawing rate: 60–120°C/sec). However such a living state of frozen protoplasm becomes unstable with increasing temperatures, as a result of recrystallization or a further development of fine ice crystals formed within the protoplasm. The recrystallization of ice in rapidly frozen tumor cells proceeds with difficulty at about ?30°C, and very easily at temperatures above ?20°C. |
