Coffee synthesizes two unique isoprenoid diterpenes, cafestol (C20H28O3) and kahweol (C20H26O3), ent-kaurene compounds produced through the mevalonate or mevalonate-independent pathway. These compounds are relevant to beverage quality, plant defense, human health and nutraceutical properties. We investigate the expression profile of genes in the early (CaHMGR, CaMVD, CaDXR and CaIDS) and final steps of isoprenoid diterpenes biosynthesis (Coffea arabica L. cytochrome P450s: CaCYP82C2, CaCYP71A25, CaCYP74A1 and CaCYP701A3) in two fruits development stages (90 and 210 days after flowering). We also observed total lipid, cafestol and kahweol accumulation in C. arabica grown using two crop systems (full sun and agroforestry) at IDR-Parana Londrina, PR, Brazil. Plants in full sun presented higher gene expression than plants grown in agroforestry systems, except for CaHMGR. Fruits showed higher levels of transcript activity 210 days after flowering, about 2.8 times more in the genes expressed during the first steps and 3.6 times in those in the later steps. Although lipid, cafestol and kahweol were higher in the full sun than the agroforestry system, only increased lipid content was statistically significant. Cultivation system plays an important role in the expression of the genes, particularly as regards light incidence. |

Prostate cancer is the most frequent and lethal neoplastic disease among men worldwide.
There is growing interest in the biologically active compounds in coffee for their potential
chemopreventive. The present study aimed to analyze the content of bioactive
compounds, the antioxidant and antiproliferative activity of green arabica coffee and
different roasts extracted by supercritical fluid. The concentration of caffeine, chlorogenic
acid and caffeic acid were evaluated. Additionally, the colorimetric profile, antioxidant
capacity, effect on cell viability, cell cycle and apoptosis of the extracts. Caffeine, caffeic
acid and chlorogenic acid content were higher in green coffee extract than in roasted
coffee (80.0mg/100g, 16.76 and 19.37mg/mL, respectively). The green coffee extract
showed higher antioxidant activity and total phenolic content compared to the other
coffee extracts by all methods tested; and among the roasted coffee extracts, light roast
coffee stands out with the highest antioxidant capacity. After 48 hours of treatment, coffee
extracts caused a reduction in the viability of PC-3 cells, with light roast coffee being
more effective. Green and light roast coffee extracts induced a cell cycle arrest in the S
phase and increased apoptosis. The present results strongly suggest that coffee extracts
have an antiproliferative effect on prostate adenocarcinoma cells.
O câncer da próstata é a doença neoplásica mais frequente e letal entre os homens em
todo o mundo. Há um interesse crescente nos compostos biologicamente activos do café
pelo seu potencial quimiopreventivo. O presente estudo visava analisar o conteúdo de
compostos bioactivos, a actividade antioxidante e antiproliferativa do café arábica verde
e diferentes torrefacções extraídas por fluido supercrítico. Foram avaliadas as
concentrações de cafeína, ácido clorogénico e ácido cafeico. Além disso, o perfil
colorimétrico, a capacidade antioxidante, o efeito na viabilidade celular, o ciclo celular e
a apoptose dos extractos. O teor de cafeína, ácido cafeico e ácido clorogénico era mais
elevado no extracto de café verde do que no café torrado (80,0mg/100g, 16,76 e
19,37mg/mL, respectivamente). O extracto de café verde mostrou maior actividade
antioxidante e teor fenólico total em comparação com os outros extractos de café por
todos os métodos testados; e entre os extractos de café torrado, o café torrado leve
destaca-se com a maior capacidade antioxidante. Após 48 horas de tratamento, os
extractos de café provocaram uma redução na viabilidade das células PC-3, sendo o café
torrado ligeiro mais eficaz. Os extractos de café torrado verde e leve induziram uma
paragem do ciclo celular na fase S e um aumento da apoptose. Os resultados actuais
sugerem fortemente que os extractos de café têm um efeito antiproliferativo sobre as
