Bright apple-green fluorescence against a dark-grey background was observed under the fluorescence microscope. The virus multiplied in monolayer cultures of lamb kidney cells. Depending upon the aggregation of virus particles, the size and brightness of the fluoresced masses became gradually prominent, and covered considerable areas of the cell cytoplasm. By indirect FAT, antibodies against SPV were detected with precision by adopting a two-stage staining procedure.
