Cassava (Manihot esculenta Crantz) lacks molecular studies for use in breeding and germplasm bank maintenance. This work aimed to develop and validate TRAP (target region amplification olymorphism) markers for cassava and evaluate their potential for structuring the genetic diversity of this species. Preliminary analyses with 396 combinations found 64%of combinations with a good amplification pattern and polymorphism. The 69 most polymorphic TRAP combinations were used to characterize 46 cassava genotypes, from which 606 alleles (range 3 to 18 with a mean of 8.8 alleles per combination) were identified. The polymorphic information content (PIC) ranged from 0.03 to 0.38 (average 0.23), while 31 combinations showed a PIC >0.25. The resolving power (Rp) parameter ranged from 0.10 to 6.30 (average 3.21). The primers that were related to starch and carotenoid biosynthesis, cyanogenic compounds, post-harvest physiological deterioration, root formation, and defense responses were the most polymorphic (>70 % of polymorphic fragments, PIC>0.25, and Rp>3.21). A total of 37 private alleles were identified in 20 accessions. Bayesian clustering as implemented in STRU CTURE revealed the presence of two major clusters (K=2) and four subclusters (K=4). The group differentiation based on the molecular variance analysis (AMOVA) showed that most of the genetic variation is within groups but with significant differences between groups. Therefore, TRAP primers have a high polymorphism for use as a molecular tool in cassava, in addition to the association with genetic regions that may increase the chances of obtaining functional markers. A antracnose, causada por Colletotrichum truncatum, é uma das principais doenças da soja. O objetivo desse trabalho foi avaliar diferentes doses de adubação potássica sobre a incidência da doença, em Sorriso/MT. O delineamento experimental foi blocos ao acaso com 6 tratamentos: 1- Adubação fixa no sulco de semeadura 350 kg/ha NPK; 2- Adubação fixa no sulco de plantio + pulverização com fungicida em V9/R1 e R.4/R5.1; 3- Adubação fixa no sulco de semeadura + 1 adubação de cobertura em V2 com 150 Kg/ha de KCl; 4- Adubação fixa no sulco de semeadura + 2 adubações de cobertura com 75 Kg/ha KCl em V2 e em V9; 5- Adubação fixa no sulco de semeadura + 3 adubações foliares com 50 Kg/ha KCl em V2, V9 e em R1; e 6 - Testemunha -lavoura comercial, e 4 repetições. Cada parcela foi constituída por 8 linhas de plantio, medindo 10 m espaçadas de 0,5 m. Foram avaliados a incidência da doença nas vagens; análise de sanidade dos grãos e produtividade. Foi utilizado o teste de Tukey a 5% de significância. Na avaliação de percentagem de antracnose nas vagens, o tratamento 1 apresentou maior percentagem da doença em relação ao tratamento 6 e este não diferiu dos demais tratamentos. Na análise de sanidade de grãos, na avaliação de Colletotrichum sp, verificou-se que o tratamento 1 apresentou percentual maior do patógeno em relação aos tratamentos 2, 3, 4 e 5 e não diferiu do tratamento 6. Não houve diferença significativa em produtividade entre os tratamentos. |
