Combined nitrogen added to nutriente media in different experiments with Phaseolus increased the dry weight of planta, percent and total nitrogen in the plant tops, but decreased the weight of nodules and the amount of acetylene reduced to ethylene by excised nodules. Nodules produced by one strain of Rhizobium demonstrated high nitrogenase activity even in treatments with nitrogen. Removal of molybdenum from culture media did not affect dry weight of plants, nodule weight, but decreased percent of nitrogen of plant tops in one experiment, while removal of magnesium decreased the dry weight of plant tops. Neither molybdenum nor magnesium deficiencies in nutrient solution influenced nitrogen fixation by different strains of Rhizobium used when measured by the acetylene reduction assay or amount of nitrogen in plant tops. Ammonium and nitrate nitrogen sources did not affect plant growth or ethylene produced by Phaseolus nodules in a growth chamber experiment. In the absence of combined nitrogen the ethylene produced by nodules was positively correlated with both total and percent nitrogen in plant tops, and with the fresh nodule weigth, while negative correlations between dry weight of plant tops and other variables (dry weight of entire plant, fresh nodule weight, and acetylene reduced) were found. In a number of soybean varieties plant age markedly influenced nodulation by increasing size of nodules, weight of nodules per plant, and influenced nitrogenase activity. The variety Wayne produced the largest nodules.... The spore ontogeny of Glomus clarum was monitored during the nonsymtiotic growth phase and after the colonization in transformed roots of clover (Trifolium repens) and carrot (Daucus carota). The germinated spore in water agar produced spore-like vesicles during the nonsymbiotic phase. It has been shown that these structures were in fact juvenile spores, that after root colonization were able to form mature spores. G. clarum began the spore formation in its nonsymbiotic phase. However, the juvenile spore structures senesced without establishment of an effective symbiosis. Spores of the genus Glomus are assumed to form terminally from sporogenic hyphae, but the ontogeny of G. clarum in transformed roots demonstrates that sporogenesis can also involve intercalary sporelike vesicles formed along the sporogenic hyphae. Using root organ culture system, all steps of spore formation can be followed in vivo without disturbing the system. This technique provides a better way to visualise events that occur during the pre- and postcolonization phase of arbuscular mycorrhiza, allowing the comprehension of full life cycle of this fungi. There are morphological variations in the ontogeny of G. clarum spores which differentiate its pattern from other Glomus species. |
