Agrobacterium-mediated transformation experiments were perments were performed with somatic embryos derived from young leaf lobes of in vitro plantlets. Four different Agrobacterium strins (EHA101, GV3111-SE, LBA4404 and ASE) were tested for their infectivity on embryogenic tissues using the uidA gene(= GUS gene) as a reporter and the nptll gene (conferring resistance to aminoglycoside antibiotics, e.g., kanamycin) as a selectable marker. The best results were obtained with the strain GV311-SE when it was grow in Minimal A medium in the presence of acetosyringone, GV3111-SE carries the plasmid pMON505 containing the above mentioned genes. In two separate experiments, GUS-expression of embryo explants was assayed after 14 day on selection medium. In both case s 5% of the explants showed dark-blue spots which were composed of more thab 10 cells. An average of 20% of the rest of the explants showed single GUS-positive cells. We believe the multicellular spots probablyderived from cell divisions of single transformed cells. These multicellular foci were generally localized on areas were secondary embryos can develop, e.g.,on the upper surface of the cotyledons. However, secondary embryogenesis has not been observed on media with kanamycin as a selective agent. Currently, experiments are under way to test the bar gene (phosphinotricin resistance gene) as a selective agent. |
