Resumo: O experimento foi conduzido com o objetivo de avaliar o efeito da adição de fibra efetiva, fibra não efetiva ou tanino em dietas contendo altos percentuais de palma sobre o consumo e a digestibilidade da matéria seca e dos nutrientes fornecidos aos ovinos. Foram utilizados quatro animais fistulados no rúmen, com peso vivo médio de 43,8 ? 1,62 kg, distribuídos em quadrado latino (4x4). Os tratamentos consistiram de uma dieta base composta por palma forrageira, milho e farelo de soja (D1) e adição de fibra efetiva proveniente do feno de tifton (D3), adição de fibra não efetiva proveniente da casca de soja (D4) e adição de ácido tânico para reduzir a fermentação ruminal (D2). O pH, a concentração de nitrogênio amoniacal e a produção total de ácidos graxos voláteis foram influenciadas pelas dietas apresentando maior produção de ácidos (175,55 ?mol/mL) e menor valor de pH (6,18) para os fluidos dos ovinos alimentados com casca de soja e maior produção de amônia (7,51 mg/100mL) para os alimentados com feno de tifton. Dietas com altas proporções de palma forrageira e com a adição de aproximadamente 15 % de uma fonte de fibra de maior digestibilidade induzem uma maior produção de AGV, porém a adição de ácido tânico diminui a fermentação ruminal.
[Parameters of fermentation ruminal of Ovines Fed with Discharges Proportions of Spineless Cactus (Opuntia fícus indicates Mill)].
Abstract: The experiment was driven with the objective to evaluating the effect of addition of effective fiber, non effective fiber or tannin to diets contends high percents of spineless cactus for ovines about intake and, dry matter and nutrients digestibility. Four canullated wethers was used, with live weight around of 43,8 ? 1,62 kg, distributed in Latin square (4x4). The treatments consisted of a diet base composed by spineless cactus, corn and soybean meal (D1) and addition of effective fiber from tifton hay (D3), addition of non effective fiber of soy peel (D4) and addition of tannis to reduce the ruminal fermentation (D2). The pH, the concentration of nitrogen amoniacal and the total production of acids volatile fat were influenced by the diets presenting larger production of acids (175,55 mol/mL) and smaller pH value (6,18) for the fluids of the ovines fed with soy peel and larger production of ammonia (7,51 mg/100mL) for fed them with tifton hay. Diets with discharges proportions of palm and with the addition of approximately 15% of a source of fiber of larger digestibility induce a larger production of AGV, however the addition of acid tannin reduce the fermentation ruminal. Guignardia citricarpa is the causal agent of Citrus Black Spot (CBS), an important disease in Citriculture. Due to the expressive value of this activity worldwide, especially in Brazil, understanding more about the functioning of this fungus is of utmost relevance, making possible the elucidation of its infection mechanisms, and providing tools to control CBS. This work describes for the first time an efficient and successful methodology for genetic transformation of G. citricarpa mycelia, which generated transformants expressing the gene encoding for the gfp (green fluorescent protein) and also their interaction with citrus plant. Mycelia of G. citricarpa were transformed via Agrobacterium tumefaciens, which carried the plasmid pFAT-gfp, contains the genes for hygromycin resistance (hph) as well as gfp. The optimization of the agrotransformation protocol was performed testing different conditions (type of membrane; inductor agent concentration [acetosyringonee - AS] and cocultivation time). Results demonstrated that the best condition occurred with the utilization of cellulose?s ester membrane; 200µM of AS and 96 h as cocultivation time. High mitotic stability (82 %) was displayed by transformants using Polymerase Chain Reaction (PCR) technique to confirm the hph gene insertion. In addition, the presence of gfp was observed inside mycelia by epifluorescence optical microscopy. This technique easy visualization of the behaviour of the pathogen interacting with the plant for the first time, allowing future studies on the pathogenesis of this fungus. The establishment of a transformation method for G. citricarpa opens a range of possibilities and facilitates the study of insertional mutagenesis and genetic knockouts, in order to identify the most important genes involved in the pathogenesis mechanisms and plant-pathogen interaction.
