O controle de nematóides gastrintestinais é realizado com anti-helmínticos, mas o desenvolvimento de populações resistentes exigiu a pesquisa de novas alternativas tais como as plantas medicinais. Existem relatos populares da atividade antiparasitária do fruto de Cocos nucifera L. Desta forma a avaliação da atividade anti-helmíntica do Liquido da Casca do Coco Verde (LCCV) e dos extratos butanólico e acetato de etila do LCCV foi realizada inicialmente através de testes de atividade ovicida e larvicida sobre Haemonchus contortus; a toxicidade aguda em camundongos foi calculada para estabelecer a dose dos testes in vivo; também foram realizados estudo fitoquímico e quantificação de taninos dos extratos e finalmente a eficácia anti-helmíntica dos extratos foi realizada através do teste controlado com o extrato acetato de etila do LCCV e para o extrato butanólico foi utilizado o teste de redução na contagem de ovos nas fezes. Foi constatada a presença de triterpenos, saponinas e taninos condensados no LCCV e nos extratos. Os teores de taninos totais no LCCV, extratos acetato de etila e butanólico foram de 19,65, 25,87 e 18,08%, respectivamente Os resultados dos testes in vitro foram promissores, no entanto os testes in vivo não demonstraram atividade anti-helmíntica contra nematóides gastrintestinais de ovinos. Abstract - The control of gastrointestinal nematodes is carried out with anthelmintics, but the development of resistant populations have required the search for new alternatives such as medicinal plants. There are popular reports of antiparasitic activity of Cocos nucifera fruit. The evaluation of anthelmintic activity of the liquid extracted from the bark of the green coconut (LBGC), butanol and ethyl acetate extracts obtained from LBGC was realized initially through the ovicidal and larvicidal activity on Haemonchus contortus; acute toxicity in mice was evaluated to establish the dosage for in vivo tests; phytochemical study and quantitative determination of total tannins level s of LBGC and its extracts were done; anthelmintic efficacy was realized using controlled test for ethyl acetate extract and for the butanolic extract the fecal egg count reduction test. Total tannins percentage found in LBGC, ethyl acetate and butanol extract was 19.65, 25.87 and 18.08% respectively. The results of in vitro tests were promising, although in vivo assays did not show anthelmintic activity against sheep gastrointestinal nematodes. Microsatellite (SSR) markers were developed for the following tropical forage species, using accessions available from the plant genetic resources (PGR) collections held by EMBRAPA (Brazilian Agricultural Research Corporation): Brachiaria brizantha, B. humidicola, Panicum maximum, Paspalum spp., Stylosanthes capitata, S. guianensis, S. macrocephala, Calopogonium mucunoides and Centrosema spp. The markers were used to analyze population structure and genetic diversity, evolution and origin of the genetic variability in the center of origin, mating systems and genetic resources in EMBRAPA?s germplasm bank. The results shed light on the amount of genetic variation within and between populations, revealed the need in some cases for further plant collection to adequately represent the species in PGR collections, allowed us to assemble core collections (subsets of the total collections) that should contain most of the available diversity and (in the case of the legumes) showed the need to avoid unwanted outcrossing when regenerating conserved material. The data will allow plant breeders to better select accessions for hybrid production, discriminate between genotypes and use marker-assisted selection in breeding programs. Our results will also underpin the construction of genetic maps, mapping of genes of agronomic interest and numerous other studies on genetic variability, population structure, gene flow and reproductive systems for the tropical forage species studied in this work.
