RFLP mapping in maize is described. Neither the order of the translocation breakpoints nor the order of the RFLP loci need be known, since presence/absence information will determine unambiguously the order of both. Correlations with the physical map are possible and a definitively ordered RFLP map can be developed - a map whose order is not based on the observation of recombinational events. Because of the numerous translocations which exist in maize, it is anticipated that the physical resolution currently exceeds the density of the RFLP map. Thus, the majority of the RFLP loci should be ordered using this technique. Currently, all 879 translocation stocks are being crossed to a standard r-x1 containing line and the appropriate tester stocks derived to provide the progeny for future screening for double hemizygotes. |
