The aim of this study was to characterize the Zn and Fe availability by phytic acid/Zn and phytic acid/Fe molar ratios, in 22 tropical maize inbred lines with different genetic backgrounds. The Zn and Fe levels were determined by atomic absorption spectrophotometry and the P through colorimetry method. Three screening methods for phytic acid (Phy) analysis were tested and one, based on the 2,2’-bipyridine reaction, was select. There was significant variability in the contents of zinc (17.5 to 42 mg.kg-1), iron (12.2 to 36.7 mg.kg-1), phosphorus (230 to 400 mg.100 g-1), phytic acid (484 to 1056 mg.100 g-1), phytic acid P (140 to 293 mg.100 g-1) and available-P (43.5 to 199.5 mg.100 g-1), and in the available-P/total-P ratio (0.14 to 0.50), Phy/Zn (18.0 to 43.5) and Phy/Fe (16.3 to 45.5) molar ratios. Lines 560977, 560978 and 560982 had greater availability of Zn and lines 560975, 560977, 561010 and 5610111 showed better Fe availability. Lines 560975, 560977 and 560978 also showed better available-P/total-P ratio. Thus, the lines 560975, 560977 and 560978 were considered to have the potential for the development of cultivars of maize with high availability of Fe and/or Zn. |

Atrazina é um dos herbicidas mais utilizados no mundo, apresentando relativa persistência em zonas saturadas e insaturadas. A principal via de dissipação de atrazina inclui a biodegradação. Solos coletados no município de Guaíra, SP, provenientes de áreas de mata, e solo sob cultivo intensivo, sob irrigação, com histórico de aplicação de atrazina, foram suplementados e incubados com o herbicida nas seguintes concentrações: 70 Mg g-1, 35 OMg-1 e 700 Mg g-1. Após diluição enm série, procedeu-se ao plaqueamento em meio de cultura (meio de Martin), também suplementado com o herbicida (700Mg g-1). Os fungos selecionados nesta fase foram cultivados em meio líquido de batata-dextrose (50%) suplementado com 700 Mg mL-1 de atrazina. As culturas foram incubadas 2 28 C, sob agitação (150rpm), por um período de 21 dias. A determinação quantitativa dos resíduos de herbicida do meio de cultura foi feita por cromatografia gasosa após extração e purificação das amostras. Verificou-se que muito embora os fungos fossem capazes de degradar o composto. Os gêneros Penicillium sp., Eupenicillium sp. e Dermatiacium sp. foram identificados como responsáveis pela degradação de atrazina mais de 90% do produto, enquanto que Penicillium sp, linhagem FG-7, degradou apenas 27%.
Atrazine is one the most widely used herbicide in the world, and is considered to be relatively recalcitrant in subsurface satired and insatured zones. The major pathways of dissipation include the biodegradation. The aim of this research was characterize a group of fungi degrading atrazine. Two types of soil (natural forest and soil with previous history of long-term application of the herbicide) coleted in north of São Paulo State, in Guaíra, were supplemented with atrazine (70, 350 and 700Mg g-1) and incubated at 28 c for 21 days. Fungi were isolated in culture medium containg, through of the serial dilution in Martin medium suplemented with the herbicide. The isolates were grown in liquid medium supplemented with atrazine in order to evaluate the capacity of degradation of each strain. nine strains belonging to the genus Penicillium, Eupenicillium and Dermatiacium grew faster, producing ligh amount of biomass using atrazine as substrate. The quantitative determination of the residues was carried out by CG after extraction and purification of the samples with ethyl acetate. All nine strains degraded atrazine with P. crustacium degrading more than 90%.
