Background - Paspalum notatum exhibits both sexual and apomictic cytotypes and, thus, is considered a good model for studies of apomixis because it facilitates comparative approaches. In this work, transcriptome sequencing was used to compare contrasting P. notatum cytotypes to identify differential expression patterns and candidate genes involved in the regulation of expression of this trait. Results - We built a comprehensive transcriptome using leaf and inflorescence from apomictic tetraploids and sexual diploids/tetraploids and a coexpression network based on pairwise correlations between transcript expression profiles. We identified genes exclusively expressed in each cytotype and genes differentially expressed between pairs of cytotypes. Gene Ontology enrichment analyses were performed to better interpret the data. We de novo assembled 114,306 reference transcripts. In total, 536 candidate genes possibly associated with apomixis were detected through statistical analyses of the differential expression data, and several interacting genes potentially linked to the apomixis-controlling region, genes that have already been reported in the literature, and their neighbors were transcriptionally related in the coexpression network. Conclusions - Apomixis is a highly desirable trait in modern agriculture due to the maintenance of the characteristics of the mother plant in the progeny. The reference transcriptome, candidate genes and their coexpression network identified in this work represent rich resources for future grass breeding programs. O uso de gelatina na composição dos meios diluidores pode aumentar a estabilidade química e física desta solução podendo vir a diminuir injúrias aos espermatozóides provocadas no processo de congelação. O objetivo desse trabalho foi avaliar os efeitos da adição da gelatina aos meios diluidores sobre a funcionalidade, viabilidade e performance reprodutiva de espermatozóides congelados de ovinos. Por meio da vagina artificial foram colhidos 24 ejaculados de seis carneiros os quais foram diluídos em meio à base de Glicina-Gema-Leite (GGL) sem (controle) e com (tratamento) adição de gelatina a 1,5%. Após congelação automatizada, as amostras do sêmen foram avaliadas quanto à: motilidade total (MT), motilidade progressiva (MP), velocidade curvilinear (VCL), velocidade em linha reta (VSL), velocidade média do percurso (VAP), amplitude do deslocamento lateral da cabeça (ALH), integridade da membrana plasmática (IMP), integridade da membrana acrossomal (IMAC) e taxa de prenhez (TP). Os resultados obtidos no sêmen controle e tratado, respectivamente, foram: MT ? 91 e 85%; MP ? 31 e 38%; VCL ? 221 e 269 μm/s; VSL ? 135 e 166 μm/s; VAP ? 173 e 209 μm/s; ALH ? 2,5 e 2,8 μm; IMP: 30 e 37%; IMAC ? 56 e 62%; TP ? 21,7e 19,6%. Conclui-se que a adição da gelatina não melhora a qualidade e fertilidade do sêmen congelado de ovinos e que, apesar dos prejuízos observados na cinética, estes não se refletiram em diminuição da fertilidade à inseminação artificial. |
