Genetic manipulation of fungi requires quick, low-cost, efficient, high-throughput and molecular tools. In this paper, we report 22 entry constructs as new molecular tools based on the Gateway technology facilitating rapid construction of binary vectors that can be used for functional analysis of genes in fungi. The entry vectors for single, double or triple gene-deletion mutants were developed using hygromycin, geneticin and nourseothricin resistance genes as selection markers. Furthermore, entry vectors containing green fluorescent (GFP) or red fluorescent (RFP) in combination with hygromycin, geneticin or nour- seothricin selection markers were generated. The latter vectors provide the possibility of gene deletion and simultaneous labelling of the fungal transformants with GFP or RFP reporter genes. The applicability of a number of entry vectors was validated in Zymoseptoria tritici, an important fungal wheat pathogen |

The giant otter, Pteronura brasiliensis, (Zimmermann, 1780), is a semiaquatic carnivore that feeds mainly on fish. Mercury has high toxicity and high potential for bioaccumulation in tissues and biomagnification in organisms through food chains. Thus, as a top predator in the trophic chain, the giant otter has the potential to accumulate mercury by biomagnification. The objective of the present study was to measure the total mercury concentration in giant otter fur samples from an area in the southern Brazilian Pantanal. Fur samples from 19 otters from different social groups were collected from captured animals or were sampled with biopsy darts. The concentrations found in the study area indicate that even in the southern Pantanal, which is approximately 475 km from gold mining activity, mercury can be considered a threat for giant otters, as well as for riverside populations. It is also important to consider that other anthropogenic sources of mercury, such as pesticides used in agriculture, could affect this population. Resumo: As ariranhas Pteronura brasiliensis são mamíferos semiaquáticos que comem principalmente peixes, cuja abundância é afetada pelas inundações sazonais e pela estrutura do habitat. Seus hábitos piscívoros levam à percepção humana negativa e conflitos com pescadores. Comparamos os hábitos alimentares de ariranhas entre estações do ano e habitats no sul do Pantanal, Brasil, analisando amostras fecais coletadas entre setembro de 2008 e junho de 2011. Investigamos se habitat e estação afetam a composição da dieta e o tamanho das presas de P. brasiliensis. Calculamos a frequência de ocorrência, frequência relativa e a sobreposição das espécies de peixes consumidas por ariranhas e os peixes capturados por pescadores. As ariranhas apresentaram um conjunto de presas mais diversificado do que os peixes capturados nas pescarias. Não encontramos uma forte sazonalidade na dieta de ariranhas, mas a composição da dieta e o tamanho das presas diferiram entre rios e lagos. A dieta das ariranhas teve maior sobreposição com os pescadores esportivos do que com os profissionais. Embora a dieta piscívora das ariranhas frequentemente leve à percepção negativa pelos seres humanos, a baixa sobreposição entre a dieta das ariranhas e as espécies capturadas na pesca local sugere que ariranhas tem um baixo efeito negativo na pesca comercial. Estes resultados indicam que programas educativos poderiam ser usados para reduzir o conflito percebido entre ariranhas e pescadores.
