To establish a strategy that would simultaneously attend the breeder's necessity of harvesting a large number of plants and the analytical capacity of the labs, the B-carotene stability during freezer storage of cassava samples was evaluated. Roots of two varieties of cassava growing in an experimental field at EMBRAPA were harvested, peeled and homogenized in a food processor. analytical sample (3-5g) were directly weighed in extraction tubes and frozen. Simultaneoosly, carotenoids of 200g of the same homogenized cassava was extracted with acetone, partitioned to petroleum ether (0,1% BHT) and collected in a volumetric flask. Aliquots were transferred to tubes, dried under nitrogen and kept at -18°C. One half of the samples were kept at Embrapa's laboratory and total carotenoid content was determined spectrophotometrically every three days. The other half was transported by airplane to UNESP's laboratory and total carotenoid and B-carotene contents were determined by spectrophotometry and HPLC, respectively. Comparison of the results from the two labs showed that the transportation time without refrigeration (12 hrs) did not affect the total carotenoid content of both samples and they were stable during 26 days. Thus, a second experiment was carried out in UNESP's lab using coaxed roots of the same varieties sent by Embrapa. Both, analytical samples and dried extracts, prepared by the same way as in the first experiment and stored at -18°C, were stable over 180 days. These results showed that the analytical samples and dried carotenoid extract can be freezer stored for later analysis. Not having HPLC, the lab can send the dried carotenoid extract to another for B-carotene determination. O desenvolvimento de cultivares do algodoeiro é uma atividade dinâmica e contínua
visando atender as demandas da cadeia produtiva da cultura, buscando obter sempre o melhor resultado. As cultivares atualmente disponibilizadas pela Embrapa Algodão para a região Nordeste possuem adaptação ao semi-árido e produtividade satisfatória, mas possuem limitações em relação à porcentagem e algumas características de fibra, principalmente a resistência. Visando suprir estas limitações e agregar mais qualidade à fibra, o programa de melhoramento genético da Embrapa está apresentando a cultivar BRS 272 ? Araripe, resultante da seleção direta aplicada sobre a cultivar CNPA
ITA 90, para tolerância à seca e adaptação ao semi-árido do Nordeste. A linha CNPA 98-2138, oriunda de seleção individual em um campo de produção de sementes genéticas conduzido no município de Touros, RN, em 1998, foi avaliada nas safras de 2002 a 2005, na rede de ensaios regionais, em 7 localidades do Nordeste brasileiro, obtendo produtividades médias superiores às cultivares BRS 8H e BRS 201 em 11,7 e 11,8%, respectivamente. Em condições irrigadas esta cultivar obteve uma produtividade média de 4679 kg/ha, superando a BRS 201 em 12,4%. Em porcentagem de fibra foi
observado um ganho de 2,6% e 0,83%, em relação às testemunhas, respectivamente. |
