Abstract: The use of reference genes is required for relative quantification in gene expression analysis and since the stability of these genes could be variable depending on the experimental design, it has become indispensable to test the reliability of endogenous genes. Therefore, this study evaluated 10 reference candidate genes in two different experimental conditions in order to obtain stable genes to be used as reference in expression studies related to scrotal hernias in pigs. Two independent experiments were performed: one with 30 days-old MS115 pigs and the other with 60 days-old Landrace pigs. The inguinal ring/canal was collected, frozen and further submitted to real-time PCR analysis (qPCR). For the reference genes stability evaluation, four tools were used: GeNorm in the SLqPCR, BestKeeper, NormFinder and Comparative CT. A general ranking was generated using the BruteAggreg function of R environment. In this study, the RPL19 was one of the most reliable endogenous genes for both experiments. The breed/age effects influenced the expression stability of candidate reference genes evaluated in the inguinal ring of pigs. Therefore, this study reinforces the importance of evaluating the stability of several endogenous genes previous their use, since a consensual set of reference genes is not easily obtained. Here, two sets of genes are recommended: RPL19, RPL32 and H3F3A for 30-days MS115 and PPIA and RPL19 for the 60 days-old Landrace pigs. This is the first study using the inguinal ring tissue and the results can be useful as an indicative for other studies working with gene expression in this tissue. Resumo: O uso de genes de referência é necessário para a quantificação relativa na análise da expressão gênica e como a estabilidade desses genes pode ser variável dependendo do desenho experimental, tornou-se indispensável testar a confiabilidade dos genes endógenos. Portanto, este estudo avaliou 10 genes candidatos de referência em duas condições experimentais diferentes, a fim de obter genes estáveis para serem usados como referência em estudos de expressão relacionados a hérnias escrotais em suínos. Dois experimentos independentes foram realizados: um com porcos MS115 com 30 dias de idade e outro com porcos Landrace com 60 dias de idade. O anel / canal inguinal foi coletado, congelado e posteriormente submetido à análise de PCR em tempo real (qPCR). Para a avaliação da estabilidade dos genes de referência, foram utilizadas quatro ferramentas: GeNorm no SLqPCR, BestKeeper, NormFinder e Comparative CT. Uma classificação geral foi gerada usando a função BruteAggreg do ambiente R. Neste estudo, o RPL19 foi um dos genes endógenos mais confiáveis para ambos os experimentos. Os efeitos raça / idade influenciaram a estabilidade de expressão dos genes de referência candidatos avaliados no anel inguinal de suínos. Portanto, este estudo reforça a importância de avaliar a estabilidade de vários genes endógenos antes do seu uso, uma vez que um conjunto consensual de genes de referência não é facilmente obtido. Aqui, dois conjuntos de genes são recomendados: RPL19, RPL32 e H3F3A para MS115 de 30 dias e PPIA e RPL19 para os suínos Landrace com 60 dias de idade. Este é o primeiro estudo usando o tecido anelar inguinal e os resultados podem ser úteis como um indicativo para outros estudos que trabalham com expressão gênica neste tecido. Abstratc: This study aimed to examine the impact of levels of dietary supplementation with fat-soluble vitamins on the production performance and egg quality of laying hens. Three hundred Hy-Line White W-36 laying hens were evaluated from 28 to 44 weeks of age. The birds were allotted to one of six treatments in a randomized block design with 10 replicates with five birds each. Performance and egg quality parameters were evaluated in four 28-day periods. A corn and soybean meal-based basal diet was formulated so as to meet the nutritional requirements of the animals, with the exception of fatsoluble vitamins. The treatments consisted of dietary supplementation with 0%, 33.3%, 66.7%, 100.0%, 133.3% or 166.7% of fat-soluble vitamins (100% supplementation consisted of 7500 IU, 2000 IU, 10 IU and 1.8 mg of vitamins A, D3, E and K per kilogram of diet, respectively). Eggshell weight, shell thickness, shell strength, feed intake, egg weight, feed conversion per egg mass and feed conversion per dozen eggs showed a quadratic response (p≤0.05) to the treatments, whereas egg mass responded linearly. Optimal results were obtained at an average fat-soluble vitamin supplementation level of 109%, which corresponds to 8175 IU of vitamin A, 2180 IU of vitamin D3, 10.9 IU of vitamin E and 1.96 mg of vitamin K per kilogram of diet. |
