To investigate the protein profiles of sugarcane exposed to drought stress, two unrelated varieties of sugarcane (Saccharum officinarum L.) RB 72910 (drought-tolerant) and RB 943365 (drought-sensitive) were subjected to progressive water stress for 30 days. RB 72910 is a non-commercial variety, but it shows high tolerance to drought and can be used as sources of genes to improve the sugarcane crop. Two physiological traits were used to quantify the water deficits in sugarcane leaves. The relative water content (RWC) in RB 943365 was decreased to 50% on day 30 whereas the RWC of RB 72910 was only slightly decreased and remained constant at 86% upon exposure to the water stress. The RB 72910 variety was able to maintain higher values of leaf water potential (?w) than the RB 943365 variety under water deficit. Water stress-related proteins were identified using a proteomics approach based on two dimensional polyacrylamide gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionizationtime of flight mass spectrometry (MALDI-TOF-MS). The expression of some proteins changed after the water deficit-treatment in RB 72910, upregulated and downregulated. While in RB 943365 the expression of all proteins decreased after treatment. An analysis demonstrated that the functions of these proteins were associated with such functions as photosynthesis, signal transduction and regulation process. These results provide insight into part of the regulatory mechanism of adaptation to water deficit through differential expression of specific proteins and highlight the potential of the variety RB 72910 for the genetic improvement of the sugarcane crop.
