Whole genome sequencing of bovine breeds has allowed identifcation of genetic variants in milk protein genes. However, functional repercussion of such variants at a molecular level has seldom been investigated. Here, the results of a multistep Bioinformatic analysis for functional characterization of recently identifed genetic variants in Brazilian Gyr and Guzerat breeds is described, including predicted efects on the following: (i) evolutionary conserved nucleotide positions/regions; (ii) protein function, stability, and interactions; (iii) splicing, branching, and miRNA binding sites; (iv) promoters and transcription factor binding sites; and (v) collocation with QTL. Seventy-one genetic variants were identifed in the caseins (CSN1S1, CSN2, CSN1S2, and CSN3), LALBA, LGB, and LTF genes. Eleven potentially regulatory variants and two missense mutations were identifed. LALBA Ile60Val was predicted to afect protein stability and fexibility, by reducing the number the disulfde bonds established. LTF Thr546Asn is predicted to generate steric clashes, which could mildly afect iron coordination. In addition, LALBA Ile60Val and LTF Thr546Asn afect exonic splicing enhancers and silencers. Consequently, both mutations have the potential of afecting immune response at individual level, not only in the mammary gland. Although laborious, this multistep procedure for classifying variants allowed the identifcation of potentially functional variants for milk protein genes. |

O estudo tem como objetivo avaliar os efeitos de um sistema de agricultura tradicional na vegetação secundária sobre a densidade e diversidade de artrópodes edáficos. A área estudada está localizada na Reserva Biológica Estadual da Praia do Sul, sudoeste da Ilha Grande, litoral sul do estado do Rio de Janeiro, onde 0,4% da reserva é utilizada para plantio de subsistência por uma comunidade de pescadores artesanais. As avaliações foram feitas, entre 1996 e 1997, em quatro momentos: capoeira, antes do plantio, 45 dias após o plantio e depois da colheita. Os resultados mostraram variações na comunidade de artrópodes edáficos ao longo do período de avaliação, evidenciando o caráter d sazonalidade dos organismos. Ao analisar a comunidade de artrópodes edáficos depois da transformação da vegetação secundária (capoeira) seguido de cultivo, foram observados efeitos drásticos sobre a densidade e diversidade na comunidade de artrópodes edáficos, pois, além da eliminação direta dos principais grupos responsáveis pela decomposição , eliminou-se, também, a serapilheira.
The study had as objective evaluates the effects in a shifting cultivation system in an secondary vegetation about density and diversity of edaphic arthropods. The studied area at located in Reserva Estadual Biologica da Praia do Sul, Southwest of the Ilha Grande, south coast of the Rio de Janeiro state, where 0,4% of the reservation are used for subsistence planting by a community of handmade fishermen. The evaluations were made, between 1996 and 1997, in four moments: secondary vegetation, before the planting, 45th days after the planting and after the crop. The results showed variations in the community structure of the edaphic arthropods in the secondary vegetation along the evaluation period, demonstrating seasonal influence to the organisms. We observed the behavior of the edaphic arthropods after the transformation of the secondary vegetation ("capoeira") followed by cultivation, and drastic effects were detected on the density and diversity of the edaphic arthropods. The principal causes were the elimination of principal saprophagous groups and also the litter.
