Resumo- Este estudo teve o objetivo de verificar o tempo de proteção do cobre contra reinfecções por helmintoses gastrintestinais de ovinos em pastejo. Foram utilizados 32 ovinos da raça Corriedale, com aproximadamente 18 meses de idade, mantidos em campo nativo naturalmente contaminado por trichostrongilideos. Os animais foram pesados e dosificados. Após, foram alocados a quatro grupos experimentais: T1 (tratado com cobre e necropsiado no 28. dia); TII (sem cobre e necopsiado no 28. dia); TIII (tratado com cobre e necropsiado no 56. dia); TIV (sem cobre e necropsiado do 56. dia). Os grupos T1 e TIII receberam via oral, uma cápsula galatinosa de quatro gramas, contendo 3,4 gramas de óxido de cobre. Os ovinos foram avaliados semanalmente através de contagem de ovos por grama de fezes (OPG), volume globular (VG), aspartato aminotransferase (AST) e concentrações de cobre no plasma. Ao abate, foram determinados: carga parasitária, concentrações de cobre no fígado e peso dos fígados. Os resultados demonstraram que os níveis de cobre no plasma não tiveram alterações significativas (P>0,05) entre os diversos tratamentos. Quanto aos níveis de cobre detectados no fígado dos animais abatidos observou-se que os lotes medicados com cobre, apresentaram níveis mais elevados que os não medicados (P<0,05), porém dentro dos parâmetros considerados não tóxicos para ovinos. Também foi observado que os níveis de AST, nos grupos tratados com cobre, apresentaram durante todo o período experimental valores levemente superiores aos dos grupos controles, mas diferença significativa (P<0,05) foi detectada apenas entre T1 e TII. Quanto à avaliação do OPG, não foram observadas diferenças significativas entre os grupos (P>0,05). Com relação aos parasitos gastrintestinais, detectou-se que a administração de cobre contribuiu significativamente para uma menor reinfestação apenas pelo Haemonchus contortus e somente nos animais abatidos na quarta semana após o tratamento, quando a redução foi de 60% nos medicados (P<0,05). Estima-se que, neste experimento, o cobre não atingiu níveis tóxicos nos animais já que não houve diferença significativa em VG, concentração plasmática de cobre, peso dos fígados e peso vivo (P>0,05). Com base nesses resultados, podemos concluir que 3,4 gramas de óxido de cobre podem ser efetivos na redução das reinfestações por H. contortus, durante quatro semanas sem causar toxicidade para ovinos criados extensivamente.
[Copper on the control of gastrintestinal nematodes in sheep].
Abstract - The aim of this work was to verify the possible activity of copper oxide wire particles against reinfection by gastrintestinal nematodes in sheep under extensive grazing. Thirty-two sheep, all wethers, aged 18 months and kept on native pastures naturally contaminated by trichostrongylides were used. The animals were weighed and treated with anthelmintics and then allocated to four experimental groups: TI (treated with copper and necropsied on Day 28), TII (untreated control and necropsied on Day 28), TIII (treated with copper and necropsied on Day 56), TIV (untreated control and necropsied on Day 56). Groups TI e TIII received, per os, a 4g bolus containing 3.4g of copper oxide wire particles. The animals were monitored, at weekly intervals, for faecal egg counts (EPG), packed cell volume (PCV), aspartate aminotransferase (AST) and plasma copper concentration. At necropsy, worm burdens, liver copper concentration and liver weights were determined. There were no differences in plasma copper levels between treatments (P>0.05). As for copper in the livers, all treated groups had higher levels than the untreated animals (P<0.05). Plasma AST activity was slightly higher during the experimental period in the treated groups. However, significant difference was only detected between treatments TI e TII (P<0.05). There were no significant differences in EPG counts between treated and control groups (P>0.05). Worm counting at necropsy revealed that copper was effective only against Haemonchus contortus and only in the animals slaughtered on Day 28º where the reduction was 60% in comparison to the untreated group (P<0.05). It is believed that in this experiment copper did not reach toxic levels as there were no significant differences in PCV, plasma copper concentrations, liver weights and body weights (P>0.05). It is concluded that 3.4g of copper oxide wire particles can have a significant action in reducing reinfections by H. contortus during four weeks without toxicity to animals under conditions of extensive grazing. The development of efficient algae cryopreservation methods is pivotal to the establishment of long-term culture collections as well as algae breeding and genetic modification programs. However, the unpredictable responses of distinct algal species to cryopreservation agents and protocols have hampered the standardization of universal methods so far. The results presented in this study indicate that intrinsic biological factors (i.e.: cell morphology and phylogenetic origin) play a role in the definition of which culture age and cryoprotectant agent type and concentration should be used to achieve successful cryopreservation. Through the use of Central Composite Rotatable Design (CCRD) it was possible to define optimized protocols for cryopreserving three major morphotypic groups found among chlorophytes (i.e.: coenobium, coccoid or palmella forming strains). These optimized protocols were then validated upon fifteen strains from Sphaeropleales, Chlamydomonadales and Chlorelalles clades. Analysis reveals that the use of DMSO as the sole cryoprotectant provided the highest postfreezing cell viability recovery rates for 80% of the coccoid strains tested, while the combination of glycerol and PEG400 allowed efficient cryopreservation of 100% of the coenobium and palmella forming strains evaluated. In addition, results suggest that coenobium-forming strains have higher resistance to freeze/thawing when frozen at mid-log phase of growth, while coccoid and plamelloid strains present higher survival rates when cryopreserved at late-log phase. Such information is of valuable practical use since a simple visual inspection of algae strain's predominant morphotype can guide the choice of the most suitable cryopreservation protocol.
