Bananas (Musa spp.) are considered the main source of income and food in many developing countries. Brazil is the second worldwide banana producer, but banana yield suffers due to the lack of commercial varieties with tolerance / resistance to main pests and diseases. One of the strategies to overcome this situation is the release of new resistant varieties through genetic breeding. Genetic linkage maps aiming the identification of alleles linked to resistance or susceptibility to biotic or abiotic stresses is an accessory tool for conventional breeding. However, for the con-struction of highly saturated genetic linkage maps, the development of suitable mo-lecular markers, such as SSRs is necessary. The main objective of the present work is to present optimized and validated microsatellites. A total of 30 primers originated from EST and BAC libraries were optimized and validated in 22 banana accessions ranging from wild to improved diploids. The number of alleles per primer varied from 2 to 11 with average of alleles per primer of 4.7. Average heterozygosity and PIC values were 0.6318 and 0.53, respectively. |
