Resumo: Treze genótipos de Urochloa mosambicensis da coleção de trabalho da Embrapa Caprinos e Ovinos, provenientes do banco de germoplasma da Embrapa Gado de Corte foram caracterizados quanto aos índices morfogênicos em casa de vegetação, durante onze meses. Os genótipos UmCO-4 e UmCO-8 apresentaram altas taxas de alongamento foliar (TAlF), aparecimento foliar (TApF) e novas folhas vivas por perfilho (NFV), favorecendo o aumento de biomassa. Os acessos UmCO-14, UmCO-4, UmCO-2 mantiveram as menores taxas de alongamento de colmo (TAlC) e maiores TAlF, favorecendo a relação folha/colmo (F/C). A média da densidade populacional de perfilhos foi de 866 perfilhos m-2, com destaque para UmCO-2 com 1.359 perfilhos m-2, tendo esse genótipo destaque por combinar aspectos favoráveis tanto para qualidade quanto potencial de forragem produzida. Os acessos UmCO-2, UmCO-4, UmCO-8, UmCO-12, UmCO-13 e UmCO-14 estão entre os mais promissores para avanço nas etapas do programa de melhoramento vegetal por seu potencial de produção e aspectos indicadores de qualidade de forragem. Abstract: Thirteen Urochloa mosambicensis ccs from Embrapa Caprinos e Ovinos working collection were characterized for morphogenic indices in a greenhouse during eleven months. The genotypes UmCO-4 and UmCO-8 showed high leaf elongation (TAlF) and appearance (TApF) rates, as well as new leaves per tiller (NFV), favoring an increase in biomass. UmCO-14, UmCO-4, UmCO-2 maintained the lowest stem elongation rates (TAlC) and and higher TAlF, favoring leaf/stem ratio (F/C). Average number of tillers population density was 866 tillers m-2, with UmCO-2 showing 1,359 tillers m-2. This genotype stood out for combining favorable aspects for both quality and high potential of forage production. UmCO-2, UmCO-4, UmCO-8, UmCO-12, UmCO-13 and UmCO-14 are among the most promising genotypes for advancing the stages of the plant breeding program due to their production potential and indicators of forage quality. Quantitative real-time polymerase chain reaction (RT-qPCR) is a powerful tool used to measure gene expression. However, because of its high sensitivity, the method is strongly influenced by the quality and concentration of the template cDNA and by the amplification efficiency. Relative quantification is an effective strategy for correcting random and systematic errors by using the expression level of reference gene(s) to normalize the expression level of the genes of interest. To identify soybean reference genes for use in studies of flooding stress, we compared 5 candidate reference genes (CRGs) with the NormFinder and GeNorm programs to select the best internal control. The expression stability of the CRGs was evaluated in root tissues from soybean plants subjected to hypoxic conditions. Elongation factor 1-beta and actin-11 were identified as the most appropriate genes for RT-qPCR normalization by both the NormFinder and GeNorm analyses. The expression profiles of the genes for alcohol dehydrogenase 1, sucrose synthase 4, and ascorbate peroxidase 2 were analyzed by comparing different normalizing combinations (including no normalization) of the selected reference genes. Here, we have identified potential genes for use as references for RT-qPCR normalization in experiments with soybean roots growing in O2-depleted environments, such as flooding-stressed plants. |
