Durante anos, muitas bactérias se tornaram resistentes a vários tipos de agentes antibacterianos, sendo de fundamental importância estudos de avaliação da resistência microbiana do sêmen frente aos agentes antimicrobianos comumente utilizados nos procedimentos de inseminação artificial. Objetivando estudar a eficiência de três diferentes antibióticos no controle de microrganismos presentes em doses de sêmen congeladas de 25 reprodutores caprinos, onde 25 ejaculados foram avaliados quanto a motilidade individual progressiva (MIP%) e vigor, após esta etapa o sêmen foi centrifugado em Tris, na proporção de 1:9 (v:v; sêmen:solução de lavagem) e, após a segunda lavagem, foi acrescentado, ao sedimento, diluente à base de leite em pó desnatado, reconstituído a 10,0 % e inativado a 95 °C, durante dez minutos. Os antibióticos foram adicionados de acordo com os grupos experimentais: A1 = penicilina + estreptomicina; A2 = gentamicina e A3 = sem adição de antibiótico (controle). Entre os dois grupos de antibióticos testados, a maior freqüência da presença de microrganismo foi registrada para Staphylococcus spp em três casos (12,0%) do total de 28,0% de microrganismos encontrados no grupo A1 ( penicilina + estreptomicina) e no A2 somente 8,0% das amostras apresentaram resistência aos microrganismos encontrados, correspondendo a Candida sp e a Shingella sonnei.
During years, many bacteria became resistant to several kinds of antibacterian agents, being of fundamental importance to undertake studies of the microbial resistance of the semen to the antimicrobians agents usually used in the procedures of artificial insemination. With the objective to study the efficiency of three different antibiotics in the microrganisms control present in frozen semen doses of 25 bucks, 25 ejaculated were evaluated regarding progressive individual motility (MIP%) and vigor, after this stage the semen was centrifuged in Tris, in the proportion of 1:9 (v:v; semen:wash solution) and, after the second wash, was added, to the sediment, diluent to the base of skimmed, reconstituted desiccated milk at 10,0 % and inativated at 95 °C, for ten minutes. The antibiotics were added according to the experimental groups: A1 = penicillin + estreptomicin; A2 = gentamicin and A3 = without antibiotic addition (control). Among groups of tried antibiotics, the biggest frequency of the presence of microrganism was registered for Staphylococcus spp in three cases (12,0%) from the total of 28,0% of microrganisms founded in group A1 ( penicillin + estreptomicin) and in A2 only 8,0% of the samples introduced resistance to microrganisms found, corresponding to Candida sp and Shingella sonnei. The presence of dormant seeds makes it difficult to evaluate physiological quality and requires the use of appropriate methods in order to break seed dormancy. The objective of this study was to evaluate the effectiveness of different methods to break the dormancy of ryegrass seeds stored under environment conditions in different periods. Ryegrass seeds, BRS Ponteio cultivar, produced in two locations were used and evaluated after 60, 90, 120, 150 and 180 days of storage under environment conditions. Seeds were submitted to the following methods to break dormancy: sowing at 20 to 30°C without applying any method to break dormancy (control); pre-cooling (5°C) for 3 days + KNO3 followed by sowing at 15 to 25°C; pre-drying (45°C) for 96 h followed by sowing at 20-30°C; immersing the seeds in sodium hypochlorite (NaClO) at 0.5% for 24 h, followed by drying at 45°C for 6 h and sowing at 15 to 25°C. At 60 days after harvest, the most effective method was immersion of seeds in a NaClO solution followed by drying at 45°C. From 90 days after harvest, all methods were equally effective to break seed dormancy, except pre-drying (45°C) for 96 h, which negatively affected the physiological quality of seeds. There is a difference in the effectiveness of the methods employed to break the dormancy of ryegrass seeds depending on the type of post-harvest storage. |
