Abstract: Cassava (Manihot esculenta Crantz) is a cianogenic species consumed as a major staple food in many tropical countries. Cianogenic glucosides are synthesized in leaf tissues tranported to starch roots where they are stored. Mechanical demage of cassava tissues leads to the release of hydrocyanic acid (HCN), a volatile highly toxic-compound. Under normal physiological conditions cassava tissues contains no detectable HCN since the cianogenic glucosides and its degrading enzymes, linamarase and hydroxynitrile lyase, exists in different in compartments. Compartmentation of linamarase has been studied previously by other authors in several species measuring enzyme activity in different tissues. In leaves, linamarase is reported to be entirely apoplastic and associated to the cell walls. However, Pancoro and Hunghes (1992) detected the expression of mRNA and the activity of linamarase only in laticifers of cassava leaves. The present study investigates the location of linamarase in leaves and roots of two varieties of cassava of low and high content of cianoglucosides by inmunoflurescent labelling. Anti-linamarase immure e serum was obtained by inoculating a rabbit with purified linamarase. Hand-cults tissues were incubated with the anti-linamarase serum and then incubated with a commercial FITC-anti-rabbit IgG conjugate. Pre-inmune serum was control. The results shows the inmunoflurescent label distributed mainly in laticifers of leaves and roots in both varieties with no differences between then. Our results supports those of Pancoro and Hughes (1992) and in addition suggest that more than one tissue mary be involved in the synthesis and transport of linamarase.
