Bacterial spot is a highly damaging tomato disease caused by members of several species of the genus Xanthomonas. Bacteriophages have been studied for their potential use in the biological control of bacterial diseases. In the current study, bacteriophages were obtained from soil and tomato leaves in commercial fields in Brazil with the aim of obtaining biological control agents against bacterial spot. Phage isolation was carried out by co-cultivation with isolates of Xanthomonas euvesicatoria pv. perforans, which was prevalent in the collection areas. In a host range evaluation, none of the phage isolates was able to induce a lytic cycle in all of the bacterial isolates tested. In in vivo tests, treatment of susceptible bacterial isolates with the corresponding phage prior to application to tomato plants led to a reduction in the severity of the resulting disease. The level of disease control provided by phage application was equal to or greater than that achieved using copper hydroxide. Electron microscopy analysis showed that all of the phages had similar morphology, with head and tail structures similar to those of viruses belonging to the class Caudoviricetes. The presence of short, noncontractile tubular tails strongly suggested that these phages belong to the family Autographiviridae. This was confirmed by phylogenetic analysis, which further revealed that they all belong to the genus Pradovirus. The phages described here are closely related to each other and potentially belong to a new species within the genus. These phages will be evaluated in future studies against other tomato xanthomonad strains to assess their potential as biological control agents. Resumo: Objetivou-se avaliar a destoxificação da torta de mamona bruta (TMB) por meio de dois produtos alcalinos em diferentes concentrações e seus efeitos sobre a composição química, degradabilidade in situ da MS e o fracionamento de proteínas. Utilizaram-se o hidróxido de cálcio [Ca(OH)2 ] e hidróxido de sódio (NaOH) em duas concentrações (60 g e 90 g), diluídos em quatro quantidades de água (1.000; 1.500; 2.000 e 2.500 ml de água por quilo de TMB). A eficácia dos tratamentos foi avaliada com base na presença das subunidades de ricina em gel a 4,19% de poliacrilamida (SDS-PAGE) em condição desnaturante por meio de análise densitométrica. Já a Ricinus aglutinina foi detectada por meio de bioensaios de hemaglutinação com hemácias de coelho. Observou-se que das diferentes concentrações utilizadas, somente a utilização de 90 g e 60 g de Ca(OH)2 e NaOH, respectivamente, conseguiu destoxificar 100% da TMB, ambas diluídas em 2.000 mL de água. Por outro lado, ao avaliar o tempo mínimo de contato dos reagentes com a TMB para uma máxima destoxificação, observou-se que, com três horas de contato, é o tempo necessário para os reagentes diminuírem em 100% as proteínas citotóxicas, além de não deixar atividade hemaglutinante nesse material. Ao avaliar a degradabilidade da MS de ambas as tortas destoxificadas (TMD), observou-se que a TMD Ca(OH)2 aumentou 10,06% da fração ?, já a TMD NaOH 22,53%. De uma forma geral, os parâmetros de degradação foram maiores para a TMD NaOH. Não houve efeito (P>0,05) das dietas sobre o desempenho dos animais, com exceção da conversão alimentar, que foi menor para as cabras alimentadas com TMD. A inclusão de TMD na dieta de cabritas em confinamento é uma opção atrativa, considerando que as cabras alimentadas com TMD apresentam melhor conversão alimentar e seu uso não provoca alterações hepáticas e renais, sugerindo que o FS pode ser completamente substituído. Abstract: The objective of this study was to evaluate the detoxification of castor cake (CC) through two alkaline products at different concentrations and its effects on chemical composition, in situ degradability of DM and protein fractionation. Calcium hydroxide [Ca(OH)2 ] and sodium hydroxide (NaOH) were used in two concentrations (60 and 90 grams) diluted in 4 quantities of water (1,000; 1,500; 2,000 and 2,500 ml of water per kg of CC). The efficacy of the treatments was evaluated based on the presence of the 4.19% polyacrylamide (SDS-PAGE) gel ricin subunits in denaturing condition by densitometric analysis. Ricinus agglutinin was detected by hemagglutination bioassays with rabbit red blood cells. From the different concentrations used, only the use of 90 and 60 grams of Ca(OH)2 and NaOH, respectively, was able to detoxify 100% of TMB, both diluted in 2,000 ml of water. On the other hand, when assessing the minimum contact time of reagents with CBC for maximum detoxification, three contact hours is the time required for reagents to reduce cytotoxic proteins by 100%. When evaluating the DM degradability of both detoxified castor cake (DCC), it was observed that DCC-Ca(OH)2 increased 10.06% of ? fraction, while DCC-NaOH increased 22.53%. In general, the degradation parameters were higher for DCC-NaOH. There was no effect (P> .05) of diets on animal performance, except to the feed conversion, which was smaller for goats fed with DCC. The inclusion of DCC in the diet of feedlot goats is an attractive option, considering that goats fed with DCC have better feed conversion and its use does not cause liver and renal disorders, suggesting that soybean meal can be completely replaced.
