Hormogaster elisae is an obligate outcrossing hermaphrodite, endemic to the central area of
the Iberian Peninsula. This earthworm species is adapted to sandy soils that are poor in organic
matter. As an endogeic and large-sized earthworm, not only is its reproductive cycle relatively
long but also its cocoon production rate is low when compared to other ecological types of
earthworms. Moreover, different amount of sperm has been observed inside its four
spermathecae, which could suggest that it is capable of separating the sperm of different partners.
As a result, sexual selection and mate choice could be expected.
When working in a cryptic system as it is the soil, molecular markers are essential to assess
mating behaviour questions. Microsatellites have been proved as a useful tool for such analysis
because of their high variability. Therefore, we successfully isolated ten polymorphic microsatellite
loci for this earthworm species. Characterization of these loci using 26 individuals revealed 8 to
25 alleles per locus and high levels of heterozygosity |
